摘 要:本文报告了一种新的心脏预适应途径即高氧预适应(hyperoxicpreconditioning,HOP)。为观察高氧预适应对鼠心肌抗氧化酶的影响,我们给大鼠吸入80%~85%氧气,1atm,每天6h,连续7d(HO组),然后进行实验并与室内空气环境下喂饲7d的大鼠(对照组)进行比较。30只Wister大鼠随机分为6组,每组5只:HO-1和对照-1组,HO吸入结束后直接处死,采用Northen杂交法测定心肌Mn-SODmRNA表达;HO-2和对照-2,采用Langendorf法造成60min缺血,60min再灌注,测心肌SOD与冠脉回流液的CPK及SOD活力;HO-3和对照-3,同法造成10min缺血,60min再灌注,测心肌与冠脉回流液的SOD活力。结果显示,HO-1组心肌Mn-SODmRNA表达明显较对照组增强。缺血及梗塞心肌SOD的活力分别为(%/g):HO-283.6±0.5;对照-276.7±1.7(P<0.01),HO-390.1±1.9;对照-385.1±1.4(P<0.05)。HO组冠脉回流液的SOD活力在缺血再灌注期明显高于对照组(P<0.001)。冠脉回流液CPK活力HO-2组再灌注30及?e report a new method of myocardial preconditioning named hyperoxic preconditionning(HOP). In order to study the effects of HOP on myocardial ischemic injury and antioxidant enzymes, the adult rats were preexposed to 80%~85% of oxygen, 1 atm, six hours per day for seven days before the experiments (HO groups). The control groups were in room air for seven days. Thirty Wister rats were devided randomly into six groups (n=5 for each group): the rats were killed afier the HO protocol and the expressions of MnSOD mRNA in myocardium were determined by Nonhern Blot (HO1 and controll); the isolated rat hearts with Langendorff method underwent 60min ischemia followed by 60min reperfusion (HO2 and control2) or 10min ischemia followed by 60min reperfusion(HO3 and control3). The activities of SOD in myocardium and in coronary efluent were measured in the last four groups,the activities of CPK in coronary efluent were measured in HO2 and control2 groups. The results showed that the expressions of MnSOD mRNA in myocardium were stronger in HOl group than that in controll group; the activities of SOD were significantly higher in HO2 and HO3 groups than those in control groups. The activities of CPK in coronary effluents were significantly lower in HO2 than those in control2 group at 30 and 60 min after reperfusion (P<0.05). Conclusion: hyperoxic preconditioning(HOP) can induce the increased expression of MnSOD mRNA and the activities of SOD in myocardium in rat.
