摘 要:花粉萌发率是检测花粉活力的一个重要指标,但实践中还缺少便捷、可靠的水稻花粉萌发率测定方法。作者在前人研究的基础上,以两系杂交中稻两优818为试验材料,采用液体培养基对相关培养条件进行了优化。结果表明,采用155 g/L蔗糖+20 mg/L H3BO3+40 mg/L CaCl2+5 mg/L V-B1相配合的培养基较适合花粉的萌发,萌发率最高可达63.78%。此外,该培养方法具有培养条件要求不高、发芽结果稳定性好、培养基分布均匀等优点。The pollen germination rate (PGR) is one of the most important factors to evaluate the pollen vltahty. Practically, low-cost, high-speed and feasible culture methods for determination of PGR are lacking. With a two-line hybrid rice cultivar LiangyouS18, we" ve tried to screen the ingredieats of a liquid culture medium for rice pollen germination test based on some previous studies. The results indicated that the culture medium with a prescription of 155 g/L sucrose +20 mg/L H3BO3+40 mg/L CACl2+5 mg/L VB1 showed significantly greater than other ingredient-based media, with a highest PGR 63.78%. In addition, this culture method was characterized by low-cost, stability in PGR, and even distribution of pollen grains on the surface of the culture medium.
