摘 要:目的探讨三碘甲状腺原氨酸(T3)对兴奋毒性脑损伤新生鼠游泳行为的影响。方法出生5d(P5)新生小鼠71只随机分为对照组、模型组和治疗组,脑内注射兴奋性脑毒素——鹅膏蕈氨酸(IA)建立兴奋毒性脑损伤模型。治疗组再分为低剂量组、中剂量组和高剂量组,并分别于造模后1h、24h、48h、72h、96h腹腔注射2μg·kg-1、5μg·kg-1、10μg·kg-1左旋三碘甲状腺原氨酸(L-T3)。设对照组,PBS替代IA和L-T3,方法同模型组和治疗组。于出生10d、12d(P10、P12)进行游泳实验,比较各组之间的差别。结果P10游泳实验总分:低剂量组、中剂量组及高剂量组与模型组比较均差异无统计学意义(Pa>0.05);P12游泳实验总分:低剂量组及中剂量组与模型组比较差异无统计学意义(Pa>0.05),但高剂量组评分高于模型组(P<0.05);P10四肢运动评分:低剂量组、中剂量组及高剂量组与模型组比较均差异无统计学意义(Pa>0.05);P12四肢运动评分:低剂量组与模型组比较差异无统计学意义(P>0.05),但中剂量组高于模型组(P<0.05),高剂量组明显高于模型组(P<0.01)。结论高剂量T3促进了兴奋毒性脑损伤小鼠游泳行为的发育。Objective To investigate the impact of triiodothyronine(T3) on swimming behavior of neonatal mice with excitotoxicity brain injury.Methods Seventy-one 5-days old(P5) neonatal mice were randomly divided into control group,model group,and treatment group.In treatment group,all pups were given intraperitoneal injection with left-triiodothyronine(L-T3) at 1 hour,24 hours,48 hours,72 hours and 96 hours after receiving intracerebral injection of 2 μg·kg-1,5 μg·kg-1 and 10 μg·kg-1 ibotenic acid (IA),subdivided into low-dese group,medium-dose group and high-dose group.In control group,IA and L-T3 were replaced with PBS to injection to the mice.Each group were compared for the differences in swimming behavior at 10 days(P10 )and 12 days(P12) after birth.Results Swimming test total scores at P10:there was no significant difference in scores between model group and low-dose group,medium-dose group,high-dose group respectively(Pa〉0.05).Swimming test total scores at P12:there was no significant difference in scores between model group and low-dose group,medium-dose group,high-dose group,respectively(Pa〉0.05),but the scores of high-dose group were higher than that of the model group(P〈0.05).Limb movement scores at P10:there was no significant difference in scores between model group and low-dose group,medium-dose group and high-dose group respectively(Pa〉0.05).Limb movement scores at P12:there was no significant difference of scores between model group and low-dose group(P〈0.05),but the scores of medium-dose group were higher than that of the model group(P〈0.05),and the scores of high-dose group were significantly higher than that of the model group(P〈0.01).Conclusion High dose T3 promoted swimming behavior of mice with the excitoxicity brain damage.
