摘 要:目的研究星形胶质细胞水通道蛋白-9(aquaporin-9,AQP9)在高渗培养液中的表达变化及AQP9在高渗性脱水中的作用。方法取生后2d的新生Wistar大鼠大脑皮层进行星形胶质细胞纯培养,分别予不同程度的高渗培养液作用于细胞,建立星形胶质细胞对高渗液反应的实验模型。通过免疫细胞化学和PT—PCR方法研究星形胶质细胞对高渗液的反应及AQIOmRNA和蛋白的表达变化。结果星形胶质细胞经渗透压为320、333mmol/L的培养液作用3、6、12、24h后,AQP9mRNA和蛋白的表达均显著高于对照组(P〈0.05),尤以作用6h后较明显,而且AQP9mRNA和蛋白的表达呈显著正相关(r〉0.7971,P〈0.01)。结论高渗引起AQP9基因表达上调,AQP9在高渗性脱水的早期可能起到重要的代偿作用。Objective To study the expression of aquaporin -9 (AQP9) and the role in cultured rat astrocytes after exposured to hypertonic medium. Methods Rat cerebral cortical astrocytes from 2 days newborn Wistar rat were undergone the pure culture and its model of cell dehydration was established by exposed to hypertonic medium. The reactivity and effect on expression of AQP9 mRNA amt protein of astrocytes to hypertonic medium were studied by RT - PCR, immunocytochemistry, and imaging analysis. Results hnmunocytochemistu- and RT - PCR analysis confirmed that the expression of AQP9 protein and mRNA was markedly increased than those in the control group (P 〈 0.05 ) after astrocytes cultured for 3, 6, 12, 24 h with hypertonic medium (320, 333 retool/L), especially expression of AQP9 was significantly increased with hypertonic medium for 6 hours. The AQP9 mRNA expression showed a positive relationship with the AQP9 protein expression when astrocytes were cultured with hypertonic medium for 1 day (r 〉 0. 7971, P 〈 0. 01 ). Conclusion The expression of AQP9 mRNA and protein increased in hypertonic medium, which indicated that the up -graduated expression of AQP9 may play an important compensation in the early stage of the hypertonic dehydration.
