摘  要:目的:观察联合应用丹参和甲基泼尼松龙对急性肺损伤(ALI)大鼠肺组织水通道蛋白-1(AQP-1)表达的影响及其机制。方法:使用内毒素(LPS)3mg/kg气管内滴入建立大鼠LPS吸入性ALI模型。50只大鼠随机分为:生理盐水对照组、LPS损伤组、甲基泼尼松龙组(LPS+甲基泼尼松龙)、丹参组(LPS+丹参)、联合用药组(LPS+甲基泼尼松龙+丹参)。观察支气管肺泡灌洗液(BALF)中蛋白含量、肺组织湿/干重(W/D)比、肺脏病理改变,免疫组化法测定AQP-1表达。结果:经气管滴入LPS可致大鼠发生ALI。与LPS损伤组相比,单用甲基泼尼松龙或丹参组BALF中蛋白含量、肺组织W/D比值均明显降低(P<0.01),肺组织AQP-1表达则明显升高(P<0.01),以联合用药组更为明显(P<0.01)。结论:联合应用甲基泼尼松龙和丹参对ALI大鼠具有良好的治疗作用,其机制可能与促进肺组织AQP-1高表达有关。Objective To investigate the effects and mechanism of combined radix salviae mihiorrhizae(RSM) and methylprednisolone on aquaporin- 1 (AQP- 1 ) expressions in lung tissues of rats with acute lung injury. Methods ALl models were induced by intratracheal instillation of LPS (3 mg/kg). Fifty rats were randomly divided into five groups: normal saline group as control, LPS group, methylprednisolone group (LPS + methylprednisolone), RSM group (LPS + RSM) and combined drug group (LPS + RSM + methylprednisolone). Protein contents in bronchoalveolar lavage fluid (BALF), the wet/dry ratio (W/D) of pulmonary tissues and the pathological changes of lungs were detected. AQP-1 expressions in lung tissues were detected by immunohistochemistry. Results Instillation of LPS (3 mg/kg) into intratracheal can induced ALI. Compared with the LPS group, in RSM group , methylprednisolone group or combined drug group, protein contents in BALF and W/D of pulmonary tissue decreased significantly(P 〈 0.01 ), while AQP-1 expression levels increased obviously (P 〈 0.01 ). Conclusion Combination of RSM and methylprednisolone has good therapeutic effects on ALl rats, and the mechanism may be in related to up-regulation of AQP-1 expressions in lung tissues.
