痰瘀同治方药脂泰胶囊抗动脉粥样硬化的机理——对c-sis、c-myc、P53基因表达的影响

摘　　要：目的观察痰瘀同治方药脂泰胶囊对家兔动脉粥样硬化相关原癌基因及抑癌基因表达的影响。方法将２４只雄性新西兰家兔随机分成４组，对照组：喂饲普通饲料。高脂组：喂饲高脂饲料。洁脂组和脂泰胶囊组喂饲饲料与高脂组相同，并分别给予洁脂２４０ｍｇ／ｋｇｄ和脂泰胶囊１．２５ｇ／ｋｇｄ。１６周末处死动物，取主动脉标本用于原位杂交。结果脂泰胶囊能逆转ｃ－ｓｉｓ，ｃ－ｍｙｃ原癌基因ｍＲＮＡ表达增强及Ｐ５３抑癌基因ｍＲＮＡ表达减弱。结论脂泰胶囊对动脉粥样硬化形成的阻抑作用与其调控原癌基因及抑癌基因表达有关。objective:To study in effects of Zhi Tai capsule on expression of oncogene c sis,c myc and antioncogene P53 in atherosclerotic lesions in rabbits.Methods:24 male New zealand rabbits were randomly divided into 4 groups.Contral group:fed with Common rabbit foods,Hypercholesterol group:fed with Hypercholesterol foods,while giving Gemfibrozil group and Zhi Tai group the same kinds of food as Hypercholesterol group with different remedies Gemfibrozil 240mg/kg·dand zhi Tai 1.25g/kg·d separatively.At the end of 16th week,the animals were sacrificed.The aortic specimens were explored with in situ hybridization.Results:The expression of c sis and c myc mRNA in atherosclerotic plaque of Hypercholesterol group were higher.However,the levels of c sis and c myc mRNA in Gemfibrozil group and Zhi Tai group were significantly lower than those in Hypercholesterol group.The expression of P53 mRNA of Hypercholesterol group was lower than that in the contral group.In contrast,the levels of P53 mRNA in Gemfibrozil group and Zhi Tai group were higher than that in the Hypercholesterol group.Conclusions:These results suggested that Zhi Tai capsule might inhibit formation of atherosclerotic plaque, which was related with regulating expression of c sis,c myc and P53 mRNA.
