摘  要:本实验将17只纯种Beagle狗分成3组,(1)胰腺炎组(P组),n=6,用2%牛磺胆酸钠加1250IU/ML胰蛋白酶的混合液,以1 mL/kg行主胰管内逆行注射,复制急性重型胰腺炎模型;(2)苦杏内酯B(BN52021)治疗组(T组),n=6,于复制急性重型胰腺炎后5min,3h分别经静脉注射血小板激活因子(PAF)受体拮抗剂BN52021(5mg/kg),(3)对照组(C组),n=5,用等量生理盐水行主胰管内逆行注射.用RM 6000多导生理记录仪测定平均肺动脉压值,8h后3组动物均以过量戊巴比妥钠静脉注射处死,立即取肺,测定肺系数(肺湿重/体重),支气管肺泡灌洗液(BALF)内细胞总数和蛋白含量,肺血管外水量/无血干肺重(EVLW/BFDL),肺组织内PAF含量.结果发现:平均肺动脉压值P组明显升高,在4h与C组、T组均有显著差异(P<0.05).P组肺系数(1.12±0.06,(?)±S,以下同),EVLW/BFDL(5.59±0.53)及BALF中细胞总数(2.03±0.20×10~9/L)和蛋白含量(44.7±6.6mg%)均显著高于C组(分别为0.94±0.04,3.85±0.20,0.90±0.09×10~g/L和23.7±1.2mg%).T组上述指标(分别为0.93±0.02,4.15±0.18,1.48±0.28X10^9/L和25.0±2.2mg%)均明显低于胰腺炎组,而与对照组相比无显著差异.肺组织内PAF含量,P组(3.76±0.39ng/g)较C组(0.78±0.1ng/g)和T组(1.41±0.12ng/g)有非常显著的增加,C组与T组之间无显?17 Beagle's dogs were divided randomly into 3 groups: (1)Pancreatitis group (PG,n = 6). The acute pancreatitis model was estabished by injecting 2% sodium taurocholate and1250 IU/ml trypsin (1 ml/kg) into the main pancreatic duct. (2) Pan+BN 52021 group (TG,n=6). Animals of TG were injected platelet activating factor (PAF) receptor antagonist BN 52021(5 ml/kg) intravenously 5 minutes and 3 hours respectively after acute pancreatitis induction. (3)Control group (CG, n = 5). Normal saline was instead injected into the main pancreatic duct.Mean pulmonary arterial pressure (MPAP) was monitored by RM 6000 physical recorder. All ani-mals were killed at 8 hours after pancreatits induction. Pulmonary vascular permeability was stud-ied by analyzing lung index (LI), protein content and cell count in bronchial alveolar lavage(BAL), and the ratio of extra vascular lung water and blood free dry lung (EVLW/BFDL). PAFcontent in lung were determined also. The results show that: MPAP of PG gradually elevated andwas significantly difference with CG and TG (P<0. 05). LI (1.2±0.06, x±s), EVLW/BFDL(5. 59±0.53),protein content (44.7±6.6 mg%) and cell count (2.03±0. 20×109/L) in BALFof PG were significantly higher than that of CG (0.94±0.04, 3.85±0. 20, 23.7±1.2 mg%, and0.90±0.09×l09/L, respectively), and TG (0.93±0.02, 4.15±0.18, 25.0±2.2 mg%, andl.48±0.28×109/L, respectively). PAF content in lung tissure of PG (3.76±0.19 ng/g) wassignificantly increased than that of CG (0.78±0.1ng/g) and TG (1.41±0.12 ng/g), The resultssuggests that: PAF receptor antagonist BN 52021 have a good effect on the pulmonary injury in-duced by acute severe pancreantitis.
