摘 要:目的:比较不同生态习性淡水鱼类对MC-LR的去毒能力与其肝脏Ⅱ相去毒酶GSTR基因诱导型表达之间的关系。方法:对实验鱼活体腹腔注射亚致死量MC-LR(50μg/kg),采用半定量RT-PCR方法,以β-肌动蛋白为外参照,测定不同生态习性淡水鱼类(如鲢鱼、尼罗罗非鱼、草鱼)肝脏8h、24hGSTR基因mRNA诱导表达水平。结果:对毒素耐受能力强的淡水鱼类(如鲢鱼、尼罗罗非鱼)8h肝脏GSTR表达升高,24h表达显著降低;而对微囊藻毒素高度敏感的淡水鱼类(如草鱼)8h、24h肝脏GSTR表达水平均无明显变化。结论:不同生态习性淡水鱼类肝脏GSTR基因诱导型表达的高低可能与其肝脏去毒能力有关,这将为深入探讨淡水鱼类MC-LR去毒相关基因的表达调控机制提供实验基础。Objective : To compare the inducible expression level of the liver phase Ⅱdetoxified enzyme Rho - class GST gene to microcystin - LR in different freshwater fishes. Methods: Using beta - actin as external control, liver Rho - class GST gene in- ducible expression levels at 8 h and 24 h among silver carp ( Hypophthalmichthys molitrix ), Nile tilapia ( Oreochromis nilotica ) and grass carp (Ctenopharyngodon ideUus ) were studied by RT - PCR, which were intraperitoneally injected with a single 50 μg/kg body weight (bwt) dose of pure MC - LR. Results: High - resistant fish (silver carp and Nile tilapia) which injected MC - LR showed significant induction( P 〈 0.05) of GSTR over controls at 8 h,while significant reduced( P 〈 0.05) at 24 h. However,in the high- sensitive fish (grass carp),there is no significant difference of GSTR gene at 8 h and 24 h.Conclusion:The difference of inducible expression level in three freshwater fishes is relative to liver detoxification ability, which would provide experimental basis on expression and regulation mechanism of microcystin - LR detoxification - related genes in freshwater fishes.
