摘 要:为了获得控制小麦籽粒大小的相关基因,本研究以小麦大粒突变体8008和烟农15为材料,利用抑制消减杂交技术,构建抑制消减cDNA文库。在双向库中随机挑取20个阳性克隆进行测序,共获得24条EST序列。在GenBank数据库中进行Blast比对和功能分析,12条EST与已知功能基因高度同源,主要包括光合作用相关基因、物质代谢相关基因、RNA功能相关基因等;5条EST序列与未知功能的禾本科作物cDNA片段高度同源,7条EST未能找到同源性匹配。In order to gain genes associated with the grain size, we employed the large grain size mutant in wheat 8008 and Yannong 15 to be as test materials, and a cDNA subtractive library was constructed by using suppression subtractive hybridization. Twenty positive clones were randomly chosen from forward and reverse libraries respectively and sequenced, a total of 24 ESTs were obtained. According to blast screening and functional annotation in GenBank database, 12 ESTs were highly homologous with the known functional genes, which could be grouped mainly into three parts, including genes associated with photosynthesis, material metabolism, and RNA function. Five EST sequences were highly homologous with unknown function of Gramineae crops cDNA fragments and 7 ESTs failed to find homology EST match. These results lay a foundation for further research on genetic of the grain weight of wheat at molecular level.
