摘 要:目的从免疫调控角度研究电针经穴对脑缺血再灌注损伤大鼠血清白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)的影响。方法采用改良线栓法制备大鼠大脑中动脉闭塞(MCAO)再灌注损伤模型,将Wistar大鼠随机分为5组,正常组、假手术组、模型组、电针非经穴组和电针经穴组,各组随机分为ld、3d、7d三个时间点,运用酶联免疫吸附实验(ELISA)方法检测大鼠血清中IL-1β、IL-6含量变化。结果与正常组比较,模型组各指标的测定结果均有显著性差异(P<0.01,P<0.05);在ld、3d时间点,与模型组、电针非经穴组比较,电针经穴组能明显降低模型大鼠血清IL-1β含量(P<0.05,P<0.01);与模型组、电针非经穴组比较,电针经穴组能明显升高模型大鼠血清IL-6含量(P<0.05,P<0.01)。结论电针经穴对脑缺血再灌注损伤大鼠血清IL-1β、IL-6的调节具有相对特异性,电针经穴通过调节免疫细胞因子IL-1β、IL-6的含量变化可能是发挥脑缺血再灌注损伤后脑保护作用机制之一。Objective To investigate, from immunoregulation, the effect of electroacupuncture at meridian points on serum interleukin-1β (IL-1β) and interleukin-6 (IL-6) in rats with cerebral ischemia and reperfusion injury. Methods A rat model of middle cerebral artery occlusion (MCAO) and reperfusion injury was made by a modified thread-occlusion method. Wistar rats were randomly allocated to five groups, normal, sham operation, model, non-meridian point electroacupuncture and meridian point electroacupuncture. Each group was randomly divided according to three time points, 1 d, 3 d and 7 d. IL-1β and IL-6 contents of rat serum were measured by enzyme linked immunosorbent assay (ELISA). Results There were significant differences in the measured indices between the model and normal groups (P〈0.01,P〈0.05). At time points 1 d and 3 d, serum IL-1β content was significantly decreased (P〈0.05,P〈0.01) and serum IL-6 content was significantly increased (P〈0.05,P〈0.01) in the meridian point electroacupuncture group compared with the model group and the non-meridian point electroacupuncture group. Conclusion Electroacupuncture at meridian points has a specific regulating effect on serum IL-1β and IL-6 in rats with cerebral ischemia and reperfusion injury. The regulation of cytokine IL-1β and IL-6 contents may be one of the mechanisms of protective action of electroacupuncture at meridian points on the brain injured by ischemia and reperfusion.
