摘 要:目的:观察针刺足三里穴对大鼠烫伤早期肠缺血和氧自由基损伤的保护作用。方法:Wistar大鼠40只,随机分为烫伤+电针组、烫伤+假针组、迷切+烫伤+电针组和迷切+烫伤+假针组,每组10只。用沸水(96℃,15 s)造成大鼠背部35%总体表面积Ⅲ度烫伤。伤后20 min开始,电针组持续针刺双侧足三里30 min,强度为2 mA,2~100 Hz;假电针组刺激非经非穴(足三里外侧旁开0.5 cm)30 min;迷切组先切断腹腔迷走神经再烫伤。于烫伤后6 h测定小肠血流量;然后处死大鼠,取肠组织测定丙二醛(MDA)含量、黄嘌呤氧化酶(XOD)和二胺氧化酶(DAO)活性及肠组织含水率(WCR)。结果:烫伤后6 h,与其余3组相比,烫伤+电针组小肠血流量和DAO活性显著增加,XOD、MDA和WCR显著降低(均P〈0.01)。与烫伤+假针组相比,两迷切组XOD和MDA水平显著增高,血流量和DAO活性显著降低(P〈0.05);两迷切组比较,上述指标的变化无统计学差别(P〉0.05)。结论:电针足三里显著增加烫伤大鼠肠黏膜血流量和DAO活性,减轻肠组织水肿和过氧化损伤,其机制可能与兴奋胆碱能通路有关。Objective:To investigate the protective effects of electro-acupuncture(EA) at Zusanli(ST36) on intestinal ischemia-induced free radical injury in rats with scald.Methods: Forty Wistar rats were randomly divided into four groups(each=10): scald+EA(S/EA),scald+shame EA(S/SEA),vagotomy+scald+shame EA(VA/S/SEA) and vagotomy+scald+EA(VA/S/EA).The full thickness scald of 35% total body surface area(TBSA) was made by 96℃ boiling water for 15 s.All animals were sacrificed after the jejunal mucosal blood flow(JMBF) was detected 6 h after scald,and the specimens of jejunum were harvested to evaluate xanthine oxidase(XOD),malondialdehyde(MDA),diamine oxidase(DAO) and the rate of water content(WCR).Results:The levels of XOD,MDA and WCR were significantly lower,and JMBF and DAO were significantly higher in S/EA group than those of other three groups after 6 h(P0.01).The levels of XOD and MDA in two vagotomy groups were significantly higher than those of S/SEA group(P0.05).There was no statistical difference between the two vagotomy groups(P0.05).Conclusion:EA at Zusanli point alleviates intestinal ischemia-induced free radical injury in rats with scald.The mechanism might be related to the cholinergic anti-inflammatory pathway.
