摘　　要：目的:研究淫羊藿总黄酮含药血清对体外培养成骨细胞增殖与分化功能的影响,进一步揭示淫羊藿"补肾壮阳,益精健骨"的作用机制。方法:采用中药血清药理学方法制备含淫羊藿总黄酮大鼠血清;用多次酶消化法体外培养成骨细胞;将含淫羊藿总黄酮大鼠血清(SRAT)以2.5%,5%,10%三种质量浓度分别加入新生大鼠颅骨成骨细胞培养液中,研究它们对细胞增殖与分化成熟的影响。细胞增殖采用MTT法进行分析;细胞分化并于培养第4、8、12、16天检测碱性磷酸酶活性(组织化学法)、Ⅰ-型胶原(Co-l)的表达(免疫组化法)和矿化结节(组织化学法)。结果:2.5%和5%SRAT表现出强烈的刺激细胞增殖活性并显著提高碱性磷酸酶活性、矿化结节数和Ⅰ-型胶原表达。结论:淫羊藿总黄酮代谢产物强烈刺激成骨细胞的增殖并促进其分化成熟,说明淫羊藿总黄酮代谢产物具有促进骨形成活性。Objective：To study the influence of serum containing epimedium flavonoids（EF） on the proliferation and differentiation of osteoblasts in vitro so as to investigate epimedium＇s mechanism with the effect of ＂reinforcing the kidney-yang,enriching essence and benefiting bone＂.Methods：Rat serum containing EF was prepared and the osteoblasts were cultured in vitro.The serum containing different doses of EF（2.5%,5%,and 10%） was added to the medium of osteoclasts.MTT assay was used to detect the proliferation of osteoclasts.Histochemical method was performed to measure alkaline phosphatase activity and mineralized nodules,and immunohistochemistry was utilized to detect typeⅠ collagen expression at the 4th,8th,12th,and 16th day.Results：EF of 2.5% and 5% strongly stimulated the cell proliferation and significantly increase alkaline phosphatase activity,numbers of mineralized nodule and type I collagen expression.Conclusion： EF might have the ability to promote bone formation.
