速效抗晕胶囊口服对Beagle犬长期毒性的研究

摘　　要：目的观察速效抗晕胶囊对Beagle犬的长期毒性。方法速效抗晕胶囊以17．5mg／kg、35．0mg、kg、87．5mg／kg剂量，连续给Beagle犬灌胃90d，停药30d，观察一般状况，称体质量，测肛温、心电图、血液学指标、血液生化指标、并做尿液检查、骨髓检查和病理组织学检查。结果与对照组相比，给药组动物服药后，随着剂量增加而更兴奋，体质量随剂量增加而下降，其余一般状况与对照组相比无明显变化；心电图、血液学指标、血液生化指标、尿液检查、骨髓检查均未见明显差异；病理组织学检查未见与药物毒性相关的主要脏器组织形态学改变；停药后给药组体质量和精神恢复正常，也未见药物延迟性毒性反应。结论速效抗晕胶囊犬长期毒性试验安全剂量为17．5mg／kg。Objective To observe the long-term toxicological effects of Suxiaokangyun capsules in Beagle dogs. Methods For long-term toxicological experiment, 24 Beagle dogs were randomly divided into 4 groups, and were given intragastrically Suxiaokangyun capsules at a dosage of 17.5 mg/kg, 35.0 mg/kg, 87.5 mg/kg for a duration of 90 days. Thirty days after termination of drug treatment, general physical status, weight, rectal temperature, ECG, hematological, biochemical and urine indices were measured and pathological examinations were also made. Results When compared with those of the control group, excitement elevated and weight of the animals decreased with increasing dosage. No statistical changes were detected in general physical status, when compared with that of the control group. And neither significant differences in ECG, hematological, biochemical and urine indices nor morphological changes probably induced by drug toxicity could be detected in the vital organs or tissues. Following termination of drug treatment, the weight and general status of the animals returned to normal, without obvious delayed drug reaction. Conclusions Animal experiments revealed that safe dosage for prolonged administration of Suxiaokangyun capsule was 17.5 mg/kg.
