Effect of the extract of Tripterygium wilfordii on hypothalamus-pituitary-adrenal axis in rats pre-treated by pentobarbital sodium

摘  要:目的研究雷公藤醋酸乙酯提取物(TWEE)对戊巴比妥钠抑制大鼠下丘脑-垂体-肾上腺轴(HPAA)的刺激作用。方法给大鼠连续14 d戊巴比妥钠(35 mg·kg^(-1),sc,bid)+TWEE(50 mg·kg^(-1),ig),同时设置对照组和模型组,分别从形态学(HE染色、Feulgen染色、FOS、JUN、PCNA和ACTH免疫组织化学染色,电镜观察)和功能(血清皮质酮、血浆ACTH检测)两方面观察TWEE对戊巴比妥钠抑制大鼠下丘脑-垂体-肾上腺轴的影响。结果戊巴比妥钠抑制下丘脑后,TWEE对肾上腺、垂体仍有一定程度的刺激作用,但TWEE对戊巴比妥钠阻断的下丘脑以及血浆ACTH和血清皮质酮没有影响。结论TWEE刺激HPAA可能是通过下丘脑或下丘脑以上的神经组织,然后再影响垂体和肾上腺;而其直接刺激垂体及肾上腺的作用较弱。AIM To study the stimulatory effect of the ethylacetate extract of Tripterygium wilfordii(TWEE) on hypothalamus-pituitary-adrenal axis(HPAA) in rats pre-treated by pentobarbital sodium. METHODS A total of 18 male Wister rats were randomly divided into three groups, control group, pentobarbital sodium group and pentobarbital sodium + TWEE group(TWEE by oral route at the same time) .To observe the effects of TWEE on HPAA in Wister rats pre-treated with pentobarbital sodium by both the function and morphology aspects, respectively. RESULTS After hypothalamus was supressed by pentobarbital sodium, TWEE still had effects of some degree of stimulating on pituitary and adrenal glands in rats,but it had no effects on hypothalamus blocked by pentobarbital sodium, ACTH in rat plasma or corticosterone in rat serum. CONCLUSION The stimulation role of TWEE on I-LPAA may mainly act by hypothalamus or more high-grade neural tissue, then affecting pituitary and adrenal glands, and the direct stimulation of TWEE on pituitary and adrenal glands is weaker.
