摘  要:目的研究磷酸锆纳米载银抗菌剂的体外抗菌效果及安全性。方法①测定不同纳米银含量的抗菌剂对金黄色葡萄球菌、表皮葡萄球菌、铜绿假单胞菌、大肠埃希菌的体外最低抑菌浓度(MIC)。②选择抗菌效果好且纳米银含量相对较低的6.0%纳米银含量的抗菌剂为实验对象,通过急性经口毒性试验、一次完整皮肤刺激试验、银离子释放试验,评价该抗菌剂的安全性。结果①3.0%、6.0%、9.0%纳米银含量的抗菌剂对大肠埃希菌、金黄色葡萄球菌和铜绿假单胞菌的MIC均为0.062 5~0.078 g·L^(-1),6.0%纳米银含量抗菌剂对多种临床分离菌株的MIC均为0.039~0.078 g·L^(-1)。②6.0%纳米银含量的抗菌剂口服毒性实验结果为实际无毒级;没有皮肤刺激性;银离子的释放量低于3.0×10^(-8) g·L^(-1)。结论①磷酸锆载银抗菌剂对多种静脉导管感染常见病原菌具有体外抗菌效果,6.0%和9.0%纳米银含量的抗菌剂均有较好的抗菌性能;选择较低浓度的6.0%纳米银含量的磷酸锆载银抗菌剂作为静脉导管材料添加剂,当6.0%的抗菌剂质量浓度在0.039~0.078 g·L^(-1)时,抗菌效果满意。②该抗菌剂的安全性能良好。AIM To study the antibacterial effect in vitro and the security of silver-containing zirconium phosphate antibacterial agents of nanometer level. METHODS(1) Minimum inhibitory concentrations (MICs) of different silver-containing antibacterial agents of nanometer level were evaluated against the strains Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa and Escherichia coll. (2) According to the MICs, the 6.0% silver-containing antibacterial agents of nanometer level were selected to study their security (such as acute oral toxicity test, dermal irritation test and release of silver ion test) because of their good antibacterial effects and relative lower silver-containing. RESULTS (1) The MICs of the 3.0%, 6.0% and 9.0% silver-containing zirconium phosphate antibacterial agents of nanometer level against Escherichia coli, Staphylococcus aureus and Pseudomonas aeruginosa were between 0.062 5 and 0.078 g·L-1. The MICs of 6.0% silver-containing one against some clinical isolates were between 0.039 and 0. 078 g·L^-1. (2) 6.0% silver-containing antibacterial agents in oral toxicity test showed they were of the actual non-toxic level, and no full skin irritation. The release dose of silver ion was less than 3.0×10^-8 g·L^-1. CONCLUSION (1)Silver-containing zirconium phosphate antibacterial agents of nanometer level have antibacterial effect against some common pathogens in the infection of venous catheter in vitro. The 6.0% and 9.0% silver-containing antibacterial agents both showed good antibacterial effects. The 6.0% silver-containing antibacterial agents are chosen as the good additives to the material of venous catheter. The antibacterial effects of the 6.0% silver-containing antibacterial agents are satisfying when the concentration is between 0.039 and 0.078 g·L- 1. (2)The security of the antibacterial agent is satisfying.
