摘  要:目的研究鼻咽癌及鼻咽癌前病变患者鼻咽组织标本基因组稳定与修复基因的表达活性特征。方法在鼻咽癌高发区中山市,筛选鼻咽炎、鼻咽癌前病变及鼻咽癌患者作为研究对象,获取鼻咽黏膜组织,应用基因组稳定和修复功能分类基因芯片(128个基因)检测相关基因表达活性,以2倍活性表达差异为比较评判标准,并以实时荧光定量PCR技术对相关基因表达活性水平进行验证。结果相比于鼻咽炎组,鼻咽癌前病变组织表达活性上调基因有22个,活性下调基因20个;鼻咽癌组织表达活性上调基因20个,活性下调基因18个。相比于鼻咽癌前病变者,鼻咽癌组织表达活性上调基因有24个,下调基因24个。乳腺癌易感基因2、Ku70结合蛋白3基因在鼻咽癌前病变及鼻咽癌组织依序呈现高表达;鼻咽癌组织表达明显下调的基因有碱基切除基因OGG1及错配修复基因MSH4、MLH3,表达明显上调的基因有RAD18;鼻咽癌前病变组织表达明显下调的基因为RAD18和泛素类基因UBE2B。结论鼻咽癌及鼻咽癌前病变患者靶器官鼻咽黏膜的基因组稳定与修复基因表达活性存在特征性表现模式。Objective To investigate the expressive characteristics of genes related to genome stability and DNA repair capacity in the local tissues of nasopharyngeal carcinoma (NPC) and nasopharyngeal precancerous lesion (NPPL). Methods Five groups of cases were chosen from Zhongshan area, Guangdong, where there is a very higher incidence of NPC, including ones of nasopharyngitis, NPPL and NPC. Nasopharyngeal tissue samples were taken from their nasopharynx respectively and gene chip techniques (dotted 128 genes per chip) were used to determine the expressive activities of genes in these tissue samples, with 2 times of expressive activity difference as the indicator for evaluation on the differentially expressed genes. Furthermore, real-time fluorescence quantitative procedures were taken to reconfirm the determined results of gene expressing activity as evaluated by gene chip techniques. Results As compared with that of nasopharyngitis, 22 genes were determined as up-regulated and 20 were evaluated as down-regulated in their expressive activities in the group of NPPL, while 20 determined as up-regulated and 18 as down-regulated in their expressive activities in the group of NPC, When compared with the group of NPPL, 24 genes were determined as up-regulated and 24 as down-regulated in their expressive activities in the group of NPC. Moreover, breast cancer gene 2 and Ku70-binding protein 3 gene were both up-regulated in nasopharyngeal tissues in an increasing tendency from NPPL to NPC in order. In NPC tissue, the most obviously down-regulated genes in their expressive activities were 8-oxoguanine DNA glyeosylase, MutS homologue gene 4 and MutL homologue gene 3, while the most significantly up-regulated one was RAD18. On the other hand, the most significantly down-regulated genes were RAD18 and UBE2B in the local tissue of NPPL.
