摘  要:目的评价重组弓形虫热休克蛋白70(rTgHSP70)联合弗氏佐剂皮下免疫小鼠的抗弓形虫感染作用。方法BALB/c小鼠70只,随机分为7组,分别为PBS组、A组(佐剂+PBS)、20PA组(20μg rTgHSP70+佐剂)、40P组(40μgrTgH-SP70)、40PA组(40μg rTgHSP70+佐剂)、60PA组(60μg rTgHSP70+佐剂)及80PA组(80μg rTgHSP70+佐剂),皮下免疫,间隔2周,加强免疫2次,共免疫3次。末次免疫后14d,用2×104个RH株弓形虫速殖子/只灌胃攻击各组小鼠,攻击后30d处死,ELISA检测血清IgG和肠液sIgA;分离脑组织内弓形虫速殖子,并计数。结果在添加佐剂组中,血清IgG水平随rTgHSP70剂量的增加而升高,与PBS组和A组比较,差异有统计学意义;肠液sIgA水平60PA组最高,与20PA和40PA组比较,差异有统计学意义;与PBS组相比,A、20PA、40P、40PA、60PA和80PA组的脑组织内速殖子数分别减少了56.87%、56.46%、61.90%、48.74%、62.87%和70.44%。结论rTgHSP70联合弗氏佐剂皮下免疫小鼠可诱导一定的抗弓形虫感染作用,80μgrTgHSP70为较佳剂量。Objective To evaluate the protective effect of recombinant Toxoplasma gondii heat shock protein 70(rTgHSP70) combined with Freund adjuvant against Toxoplasma infection in mice. Methods Seventy BALB / c mice were randomly divided into PBS, A(adjuvant + PBS), 20PA(20 μg rTgHSP70 + adjuvant), 40P(40 μg rTgHSP70), 40PA(40 μg rTgHSP70 + adjuvant), 60PA (60 μg rTgHSP70 + adjuvant)and 80PA(80 μg rTgHSP70 + adjuvant)groups, immunized for 3 times at intervals of 2 weeks, and challenged intragastrically with 2 × 104 tachyzoites of Toxoplasma gondii RH strain 14 d after the third immunization. All the mice were killed 30 d after challenge and determined for IgG in sera and sIgA in intestine washes by ELISA, and the Toxoplasma gondii tachyzoites in their brain tissues were isolated and counted. Results The serum IgG levels of mice in 20PA, 40PA, 60PA and 80PA groups increased with the increasing dosages of rTgHSP70, which showed significant difference with those in PBS and A groups. The sIgA level in intestine washes of mice in 60PA group was significantly higher than those in 20PA and 40PA groups, which was the highest in the seven groups. Compared with that in PBS group, the counts of Toxoplasma gondii tachyzoites in brain tissues of mice in A, 20PA, 40P, 40PA, 60PA and 80PA groups decreased by 56. 87%, 56. 46%, 61. 90%, 48. 74%, 62. 87% and 70. 44% respectively. Conclusion Immunization with rTgHSP70 combined with Freund adjuvant by subcutaneous injection induced a certain protective ef-fect against Toxoplasma infection in mice, and the optimal dosage of rTgHSP70 was 80 μg.
