摘 要:目的:探讨慢性肾功能衰竭中医不同证型的血清蛋白质谱的变化及生物学基础。方法:收集慢性肾功能衰竭患者83例及正常人36例并辨证分型,采用SELDI-TOF蛋白芯片技术对不同证型的血清蛋白质表达谱进行研究。结果:在慢性肾衰中医各证型的整体研究中,17个蛋白峰有显著差异(P<0.001),通过层次聚类分析显示中医各证型之间聚类性质差异较小;通过对中医不同证型作两两比较发现,脾肾气虚组与脾肾阳虚组之间得到2个差异蛋白峰(P<0.01),脾肾气虚组与阴阳两虚组得到4个差异蛋白峰(P<0.01),脾肾气虚组与脾肾气阴两虚组之间得到5个差异蛋白峰(P<0.01),脾肾阳虚组与阴阳两虚组之间得到4个差异蛋白峰(P<0.01),脾肾阳虚组与脾肾气阴两虚组之间得到4个差异蛋白峰(P<0.01),阴阳两虚组与脾肾气阴两虚组之间得到2个差异蛋白峰(P<0.01);运用PCA分析显示,各证型组之间的差异蛋白峰能较好把各组相区分。结论:采用SELDI-TOF-MS蛋白芯片技术分析慢性肾功能衰竭中医不同证型血清蛋白质谱变化对探讨中医证型的生物学基础具有一定的意义。Objective:To explore the changes in serum protein spectrum and biological foundation of different TCM syndrome types of Chronic renal failure(CRF).Methods:Collecting 83 patients of CRF and another 28 normal cases as controlled group and differentiating the TCM syndrome to study serum protein spectrum of different TCM syndrome types of CRF by SELDI-TOF.Results:A total of 17 effective protein peaks were significantly different between TCM syndrome types and healthy subjects(P0.001)and it was smaller difference from the nature of the clustering in different TCM syndrome types.By comparison from different TCM syndrome types,there had 2 effective protein peaks(P0.01)between Spleen and kidney qi deficiency group and Spleen and kidney yang deficiency group,4 ones(P0.01)between Spleen and kidney qi deficiency group and Yin and yang deficiency group,5 ones(P0.01)between Spleen and kidney qi deficiency group and Spleen and kidney qi and yin deficiency group,4 ones(P0.01)between Spleen and kidney yang deficiency group and Yin and yang deficiency group,4 ones(P0.01)between Spleen and kidney yang deficiency group and Spleen and kidney qi and yin deficiency group,2 ones(P0.01)between Yin and yang deficiency group and Spleen and kidney qi and yin deficiency group;Protein peaks of different TCM syndrome types can better distinguish the various groups by analysis of PCA.Conclusion:Analysis of serum protein changes of different TCM syndrome types of CRF by SELDI-TOF-MS protein chip technology could have a significance on research to biological foundation of different TCM syndrome types.
