摘  要:目的探明转录因子c-Myc在同步化之后的小鼠毛囊周期中表达的规律及可能意义。方法通过经典的松蜡合剂脱毛法使小鼠背皮毛囊周期同步化,分别采用免疫组织化学、RT-PCR、Western blot检测c-Myc在毛囊周期不同时相点(拔毛后5、10、16、20、25d)表达的强弱变化及分布规律。结果在所有5个时相点的皮肤中c-Myc都存在不同强度的表达,RT-PCR与Western blot检测结果呈现出一致的规律,即生长期中期(10d)、晚期(16d)和退化期(20d)表达较强,生长期早期(5d)和静止期(25d)表达较弱。免疫组织化学结果显示:c-Myc表达在生长期主要分布于毛囊内根鞘与毛母质,在退化期主要分布于残存的内根鞘,在静止期毛囊中没有表达。结论c-Myc的表达存在于生长期的内根鞘、毛母质和退化期残留的内根鞘。c-Myc可能在毛囊角质形成细胞的终末分化过程中发挥重要作用。Objective To detect the expression pattern of transcription factor c-Myc in synchronized hair follicles in mice at different hair follicle stages and explore its possible significance. Methods Pine wax mixture to removal hair shafts was used to synchronized mouse back skin hair follicles. Totally 15 female C57BL/6 mice (hair follicles in the anagen phase) were randomly divided into 3 groups for immunohistochemieal staining (IHC) , RT-PCR and Western blot analysis respectively. The mice were killed in 5, 10, 16, 20 and 25 d after the infliction. The intensity and localization of c-Myc in different hair follicle stages were detected. Results The expression intensity of c-Myc were different in all 5 time points in the skin. The results of Western blot analysis and RT-PCR are consistent, showing that c-Myc was strongly expressed in the middle and late anagen phase and in the catagen phase while weakly in the early anagen and telogen phases. IHC results showed that c-Myc was mainly distributed in the inner root sheath (IRS) and the matrix in the anagen phase, and rudimentary IRS in the catagen phase. There was no expression in the telogen phase. Besides, c-Myc expression was strong in the sebaceous glands in all hair follicle stages. Conclusion c-Myc is specifically localized in the IRS and matrix during the anagen phase, and the rudimentary IRS in the catagen phase. Our findings reveal that c-Myc may play an important role in terminal differentiation of hair follicle keratinocytes.
