摘 要:目的:观察高糖环境下豚鼠膀胱Cajal间质细胞(interstitial cells of Cajal,ICCs)超微结构,探讨糖尿病膀胱(diabetic cystopathy,DCP)的发病机制。方法:采用酶消化法原代培养,透射电镜观察5、10、15mol/L葡萄糖浓度下培养24、72h的ICCs超微结构。结果:随糖浓度增高培养时间延长,ICCs内细胞器明显减少,线粒体大小不等,数量减少,肿胀、空泡样改变甚至溶解,内质网扩张、空泡样变,胞质广泛溶解,突起消失。结论:高糖环境对豚鼠膀胱ICCs超微结构造成显著影响,这种结构破坏可能是造成DCP发病的重要原因之一。Objective:To investigate the ultrastructural of guinea pig bladder Cajal interstitial cells(interstitial cells of Cajal, ICCs)in the high glucose environment,and to investigate the mechanism of diabetic cystopathy(DCP).Method:Primary culture ICCs bladder cells was substitute by collagenase digestion.The thultrastructural of ICCs at different concentrations(5,10,15mol/L)and different times(24,72h)were observed by transmission electron microscope.Results:The organelles in ICCs declined with the glucose concentration and culture time increased.The mitochondrion were inequality of size,abated of quantity,swelling and distension,cavity affected and even dissolution.Endoplasmic reticulums were extension,cavity affected.Cytoplasm were deliquesced generally.Scabrosity vanished.Conclusions:High glucose environment significantly affected the the ultrastructural of ICCs,which would be one of the most important etiological factor for DCP.
