摘  要:目的:观察大鼠实验性脑出血后神经细胞的凋亡及胶质纤维酸性蛋白(glial fibrillary acidic protein,GFAP)的表达。方法:将成年SD大鼠随机分为正常组、假手术组和脑出血组。脑出血组大鼠通过立体定向术向脑内注入自体动脉血制成脑尾壳核出血模型,并按不同的再喂养时间(1、3、7、14及30d)分为5个亚组。采用Hoechst和免疫组织化学染色分别观察各组大鼠出血灶周边区神经细胞凋亡和GFAP的表达。结果:与正常组和假手术组相比,脑出血组大鼠的细胞凋亡数显著增高(1~30d时P<0.05),并在3d时达高峰;脑出血1dGFAP表达开始增加,7d达高峰,以后逐渐下降,但30d时仍明显高于正常组和假手术组。结论:脑出血可诱导神经细胞凋亡,同时也促进出血灶周边区星形胶质细胞增生,这可能是脑出血后神经组织重塑的重要方式。Objective: To observe the apoptosis of neuronal cells and the expression of GFAP in brain tissue of rats following experimental intracerebral hemorrhage. Methods:The adult SD rats were randomly divided into normal group, sham group and intracerebral hemorrhage group. On rats of intracerebral hemorrhage group autologous arterial blood was stereotaxically injected into the into 5 subgroups according to the refeeding time ing methods were introduced to observe the apoptosis caudate-putamen nucleus. Then the rats were subdivided 7, 14 and 30 d). Hoechst and immunohistochemical stainof neuronal ceils and the expression of GFAP. Results: Compared with normal group and sham group, apoptosis cells in rats of intracerebral hemorhage group were upregulated(P〈0.05 on 1d-30d), and reached the peak at 3rd d. The number of GFAP-positive cells increased at the 1st d after intracerebral hemorrhage, and reached the peak at the 7th d. From then on, it decreased gradually, although the numbers of PCNA-positive cells in intracerebral hemorrhage group at the 30th d was still significantly increased compared with those of the normal group and sham group. Conclusion : Intracerebral hemorrhages stimulates apoptosis of neuronal cells and proliferation of astrocyte, and these may play an important role in neural tissue rebuild after intracerebral hemorrhage.
