摘 要:[目的]采用紫外诱变的方法选育谷氨酰胺合成酶高产菌株,并对其酶学性质进行研究。[方法]以紫外诱变选育谷氨酰胺合成酶产生菌Arthrobacter globiformis A0014,通过测定其谷氨酰胺合成酶酶活力,获得高产突变株,之后进一步对其酶学性质进行研究。[结果]获得高产突变株Y3。该菌株经多次传代,遗传性状稳定,其谷氨酰胺合成酶摇瓶发酵单位是出发株的2.81倍。突变株谷氨酰胺合成酶最适反应温度为40℃,最适pH值为6.5。[结论]所获得的突变株酶活的提高以及最适反应温度的下降有利于酶法合成谷氨酰胺,在工业生产中将有较好的应用前景。[Objective]The research aimed to breed the high-yield strains of Glutamine synthetase(GS) by UV mutagenesis and study its enzymatic properties.[Method]A mutant strain with high GS productivity were obtained through treating the Arthrobacter globiformis A0014 by UV mutagenesis,and its enzymatic properties were studied.[Result]The mutant strain with high GS productivity was Y3.The high GS productivity of the mutant strain could be inherited after repetitious subcultures.Its genetic properties was stable,and activity was 2.81 times of of the original one.The GS's optimal reaction temperature and pH value of the mutant strain were 40 ℃ and 6.5,respectively.[Conclusion] The high enzyme activity and the low optimal reaction temperature of the mutant strain obtained by UV mutagenesis is in favor of enzymatic synthesis of L-glutamine,and it will have a good prospect in industrial production.
