摘  要:目的观察宝灵膏对脑缺血再灌注损伤大鼠Caspase-3表达的影响。方法采用双侧颈总动脉阻断的方法建立大鼠脑缺血再灌注损伤模型,随机分为假手术组、模型组和宝灵膏高、中、低剂量组。采用连续切片免疫组化技术检测脑组织中Caspase-3表达的部位及数量。结果宝灵膏高、中剂量组皮层、纹状体Caspase-3免疫阳性细胞数与模型组比较有统计学差异(P<0.01),与低剂量组比较有统计学差异(P<0.05)。结论宝灵膏可以下调沙鼠脑缺血再灌注损伤后脑组织皮层和纹状体Caspase-3的表达,从而起到一定的脑保护作用,并且具有剂量依赖性。Objective To explore the effect of Bowring Paste (BP) on expression of caspase-3 after cerebral ischemia reperfusion injury (IRI) in mongolian gerbil. Methods Rats were made cerebral ischemia-reperfusion injury with bilateral carotid artery occlusion method and then randomly divided into sham group, model group, bowring paste treatment groups (high-dose, medium-dose, low- dose). A continuous sections immunohistochemical technique was applied for detection of the location and quantity of Caspase-3 expression in brain tissue. Results Bowring paste high-, medium-dose treatment groups were observed statistical significance in comparison with the model group on Caspase-3 positive cells in cortex, striatum(p〈0. 01), also with low-dose group(p〈0. 05). Conclusion Bowring paste can reduced the expression of Caspase-3 in cortex and striatum of brain tissue after cerebral ischemiareperfusion injury in mongolian gerbil, which plays a dose-dependent role in brain protection.
