摘 要:目的比较大理猪源旋毛虫(Trichinela spiralis)成虫与肌幼虫表面抗原(Surface antigen,SA)的蛋白组分差异,并进一步分析其免疫反应性,为分离和筛选出免疫原性和免疫反应性强的旋毛虫抗原成分提供依据。方法采用十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE),对成虫和肌幼虫SA蛋白成分进行分析,并进一步采用蛋白印迹(Western-blot)对其免疫反应性分析比较。结果旋毛虫成虫SA显示4条主要蛋白染色条带,分子量分别为50kDa、48kDa、40kDa和34kDa,旋毛虫肌幼虫SA显示10条主要蛋白染色条带,分别为105kDa、99kDa、80kDa、63kDa、55kDa、48kDa、35kDa、28kDa、24kDa和21kDa。;Western-blot检测结果表明,旋毛虫成虫SA出现4条反应条带,其分子量分别为50kDa、48kDa、40kDa和34kDa,旋毛虫肌幼虫SA出现7条反应条带,分子量分别为105kDa、99kDa、80kDa、55kDa、48kDa、35kDa和28kDa,其中48kDa和35kDa显色明显。结论旋毛虫成虫SA与肌幼虫SA都含有分子量为48kDa的蛋白组分,且两种抗原组分均具有较强的免疫反应性。Objective To comparatively analyze the protein components and the immunoreactivity of the surface antigens of Trichinella spirolis adult worm and muscle larvae in Dali. Methods The protein components and immunoreactivity of the surface antigens of Trichinella spiralis adult worm and muscle larvae were analyzed by using sodium dodecyl sulfatepolyacryl amine gel eleetrophoresis (SDS-PAGE) and immunoblotting test (Western-blot). Results The surface antigens of T.spiralis adult worm displayed 4 main protein dyeing bands by SDS-PAGE,and the molecular weights were 50kDa, 48kDa,40kD, 34kDa respectively; and displayed 4 reaction bands by Western-blot , the molecular weights were 50kDa,48 kDa,40 kDa,34kDa respectively. The surface antigens of T.spiralis muscle larvae had 10 main protein dyeing bands ,and the molecular weights were 105kDa,99kDa,80kDa,63kDa,55kDa,48kDa,35kDa,28kDa,24kDa,21kDa;the surface antigens of T.spiralis muscle larvae displayed 7 reaction bands and the molecular weights werel05 kDa,99 kDa,90 kDa,55 kDa,48kDa,35kDa and 28kDa,in which the reachtion bands of 48kDa and 35kDa were colored deeper. Conclusion T.spirdis adult worm and muscle larvae have common protein component sand the molecular weight is 48kDa, and both the surface antigens of adult worm and muscle larvae have strong immunoreactivity.
