摘  要:目的观察血红素氧合酶-1(HO-1)及基质金属蛋白酶-9(MMP-9)蛋白在兔主动脉粥样硬化病变的表达及普罗布考对HO-1、MMP-9表达的影响。方法16只新西兰大白兔建立动脉粥样硬化症模型后,随机加喂普罗布考(n=8,普罗布考组)或淀粉(n=8,淀粉组)6周。检测两组兔以及正常新西兰大白兔(n=8,正常对照组)血脂、体重和血清丙二醛含量。采用免疫组化法测定主动脉HO-1以及MMP-9蛋白的表达。结果与淀粉组相比,普罗布考组动脉粥样斑块缩小,HO-1在斑块内及动脉平滑肌内表达增多,MMP-9在斑块内表达减少,血清总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)、高密度胆固醇(HDL-C)和丙二醛明显降低(P均<0.05)。结论普罗布考在降低胆固醇的同时,还可能通过上调HO-1、下调MMP-9发挥作用,这可能是其抗动脉粥样硬化的重要途径。Objective To observe the effect of probucol on the expression of heme oxygenase-1 (HO-1)and matrix metalloproteinase-9 (MMP-9) protein in atherosclerotic lesions in rabbits. Methods The hypercholestrolemic animal model was estabhshed by 8 week's high cholesterol diet. Then 16 New Zealand rabbits were randomly fed with starch (n=8, starch group ) or probucol (n=8 ,probucol group ) for 6 weeks. The level of serum lipid,MDA.and body weight of rabbits in starch group,probueol group and control group (n=8, control group )were determined. The expressions of HO-1 and MMP-9 were detected by immunohistochemistry. Results The atherosclerotic plaque was shrunk and the level of HO-1 decre3ased in probucol group. The expression of aorta smooth muscle cell ineeased and the level of MMP-9 in atheroselerotic plaque reduced. The levels of down-regulated serum total cholesterol (TC),low-density lipoprotein-cholesterol (LDL-C), high-density lipoprotein-cholesterol (HDL-C) and malon dialdehyde (MDA) were significantly decreased (P〈0.05). Conclusion While lowering level of serum cholesterol, probucol possesses anti-atherogenic actions through up-regulation of HO-1 expression and down-regulation of MMP-9 expression.
