摘 要:目的观察邻苯二甲酸二(2-乙基已基)酯(DEHP)对大鼠睾丸能量代谢酶的影响及其氧化损伤作用。方法将32只健康雄性大鼠随机分为4组,即阴性对照组(玉米油)和DEHP染毒组(375、750、1500mg.kg-1)。采用经口灌胃的方式重复染毒30d,于末次染毒24h后,取右侧睾丸制备组织匀浆,采用分光光度法测定LDH、SDH、ATPase及SOD、MDA、GSHPx和GSH的活性。结果随着染毒剂量增加,与对照组比较,SOD活力与MDA含量增高,1500mg.kg-1组差异具有统计学意义(P<0.05);GSH与GSH-Px活力先增高后降低,GSH-Px活力差异均具有统计学意义(P<0.01),GSH活力差异均无统计学意义(P>0.05);SDH酶活性增高,差异均具有统计学意义(P<0.01);Na+-K+-ATPase的活性750mg.kg-1组差异具有统计学意义(P<0.01),LDH、Ga2+-ATPase、Mg2+-ATPase活性差异均无统计学意义(P>0.05)。结论DEHP可能通过影响睾丸生殖细胞能量代谢及脂质过氧化损伤作用对雄性大鼠睾丸发挥毒性作用,其毒作用特点可能表现为毒物兴奋效应。Objective To observe the effect of phthalate (2-ethylhexyl) ester(DEHP) on rat testicular enzymes of energy metabolism and its role in oxidative damage. Methods 32 healthy male rats were randomly divided into four groups, namely the negative control group (corn oil) and DEHP exposured group(375,750,1500mg·kg^-1). Using oral gavage repeat exposure 30d, at 24h after the last exposure, the right side testicular tissue were taken and prepared. LDH, SDH, ATPase and SOD, MDA, GSHPx and GSH activity were determined using spectrophotometrie method. Results Compared with the control group, As the dose increased SOD activity and MDA content increased and the high-dose group showed significant differences (P 〈 0.05); vitality of GSH and GSH-PX increased after the first reduction; GSH-PX vitality showed significant differences (P 〈 0.01 ) between groups; SDH activity increased, the differences had statistically significant( P 〈 0. 01 ) ; Na^+ -K^+ - ATPase activity in the middle-dose group had a statistically significant difference( P 〈 0.01), LDH, Ga^2+ -ATPase, Mg^2+ -ATPase activity had no statistically significant difference ( P 〉 0.05). Conclusion DEHP may play a role in toxicity of the testis of male rats by affecting the testicular germ cell energy metabolism and lipid peroxidation damage activity.
