摘  要:目的了解福州地区人博卡病毒(HBoV)在儿童呼吸道感染中的检出情况,并对其进行全基因组序列测定和种系分析。方法收集2007年11月至2008年10月在福建省妇幼保健院因下呼吸道感染住院的重症监护病房的57例小儿鼻咽抽取物标本,用一对特异引物通过PCR扩增法对HBoV基因片段进行检测,对检测出的2例HBoV(FZ1和FZ40)用7对全序列引物进行扩增和拼接,获得这两株病毒全基因组序列,上传GenBank并与基因库中国内外其它10株HBoV的全基因组序列和各氨基酸序列进行比对,并做种系分析。结果FZ1株基因组序列全长为5299bp,与HBoV参考株st2株序列长度相同;而FZ40株的基因组序列全长少2bp。病毒全基因组编码4种蛋白,分别是非结构蛋白NS1、核蛋白NP-1和衣壳蛋白VP1、VP2。结论种系分析显示福州的FZ40株与浙江温岭的WLL-1株关系较近,而FZ1株与北京的两株及泰国的CU6株关系较近。The human boeavirus (HBoV) most likely the second known parvovirus species pathogenic to humans was investigated. In this study, the presence of HBoV in children with respiratory tract disease in Fuzhou, Fujian, China was detected. Nasopharyngeal aspirates were collected from children with respiratory tract disease from Nov. 2007 to Oct. 2008. A total of 57 clinical samples were tested for HBoV using PCR assay. The complete genome of the two HBoV isolates (FZ1, FZ40) were sequenced and deposited in GenBank (Acession Nos. GQ455987, GQ455988). It was demonstrated that the length of two sequences of the HBoV isolates in this study varied each other, and the sequence length of strain FZ1 in nucleotides was 5 299bp, which was 2bp longer than that of FZ40. The deleted base pair located at site 5 286 in the end of the non-coding region. The complete genome of the HBoV encoding 4 proteins and they were non-structural protein NS1, nuclear protein NP1 and two capsid proteins VP1, VP2. We had compared the complete genome sequence of the two HBoV isolates in Fuzhou, China to other HBoV isolates and other virus of Parvovirinae documented in the GenBank database by using PHYLIP software version 3.65 and the neighbo〉joining method. The result of phylogenetie anaiysis showed that the 2 HBoV strains in our study were not clustered together and strain FZ40 was more closed to the WLL-1 strain of China than strain FZ1. Meanwhile, the strain FZ1 was more closed to the two strains of Beijing and CU6 strain of Thailand.
