摘  要:目的研究白藜芦醇(Res)逆转人乳腺癌MCF-7/阿霉素(ADM)细胞多药耐药(MDR)的作用及初步机制。方法采用MTT法测定不同浓度Res对MCF-7/ADM细胞的抑制作用、应用金氏公式判断Res和ADM联合效应;同时采用荧光分光光度法对不同浓度Res作用下MCF-7/ADM细胞内的ADM积量进行测定。结果Res能提高MCF-7/ADM细胞对ADM的敏感性,ADM联合4μmol/L Res时逆转倍数为1.950,8μmol/L为2.178,12μmol/L为2.355;Res可提高ADM在MCF-7/ADM细胞内的蓄积,耐药细胞MCF-7/ADM内ADM的浓度明显升高(与对照组比较,P<0.05),且细胞内ADM浓度均随细胞外ADM浓度升高而升高。结论Res对ADM抗人乳腺癌MCF-7/ADM细胞具有增敏作用;Res能够部分逆转MDR,其逆转机制与增加ADM药物浓度有关。Objective To study the reversal effect of resveratrol on multidrug-resistanee of human breast cancer cell line MCF-7/ADM, and the related mechanism. Methods MTT assay was used to detect the inhibitory effect of resveratrol of different concentrations on the proliferation of human breast cancer cell line MCF-7/ADM, and the Jin's formula to assess the combined effect of Res and ADM on cancer line MCF-7/ADM. In addition, the concentrations of ADM in MCF-7/ADM cells were detected by Fluorescence Speetrophotometry while Resveratrol of different concentrations was given. Results Resvera- trol combined with ADM possessed synergistic effect, and the reversal index was 1. 950 in ADM +4 μmol/L Res, 2. 178 in ADM+ 8 μmol/L Res, and 2. 355 in ADM+ 12 μmol/L Res. Resveratrol was observed to greatly elevate the concentration of ADM in MCF-7/ADM cells at a dose-related manner (com- pared to the control group, P〈0.05). Conclusions Resveratrol has a sensitizing effect on multidrug-resistance of human breast cancer cell line MCF-7/ADM in that it could partially reverse MDR in MCF-7/ ADM cells. The reversal is coorelated with elevated concentration of ADM in MCF-7/ADM cells.
