摘  要:目的观察辛伐他汀对骨质疏松大鼠骨代谢和股骨生物力学的影响。方法将SD大鼠随机分成假手术组(SHAM)、卵巢切除组(OVX)、卵巢切除加辛伐他汀组(OVX+SIM),每组16只。适应性喂养1周后手术,术后8周给药,OVX+SIM组给予辛伐他汀5 mg.kg-1.d-1,其余2组用等量生理盐水灌服。用药后第4、12周分2批各处死半数大鼠,取血观察各组大鼠的骨源性碱性磷酸酶(BALP)、骨钙素(BGP)的代谢情况。分离股骨进行三点弯曲试验,检测股骨最大载荷、弹性载荷、最大桡度、弹性桡度等生物力学性能。结果(1)用药4周,BALP水平OVX+SIM组较OVX组升高(P<0.05),弹性载荷OVX+SIM组较OVX组降低(P<0.05)。(2)用药12周,BALP水平、BGP水平OVX+SIM组较OVX组升高(P<0.01),最大载荷、弹性载荷OVX+SIM组较OVX组增加(P<0.05,P<0.01)。(3)用药12周与用药4周时比较,OVX+SIM组BALP水平降低(P<0.01),OVX+SIM组BGP水平升高(P<0.01),OVX+SIM组最大载荷、弹性载荷均有明显增加(P<0.01)。结论辛伐他汀显著影响去势大鼠的骨代谢,促进成骨,改善骨生物力学性能。Objective To investigate the effect of simvastain on bone metabolism and bone biomechanical characteistics of femur vertebra in rats with ovariectomy-induced osteoporosis. Methods Fortyeight female SD rats were divided by randomized block design into three groups: the sham-operated group (SHAM), the ovariectomized group (OVX), and the simvastatin ovariectomized group (OVX+SIM). Eight weeks after the operation, simvastatin was administered orally with 5 mg·kg^-1·d^-1 to the OVX+ SIM group and normal saline to the other two groups. The rats were killed separately 4 weeks and 12 weeks after the administration. Blood samples were collected to obtain serum levels of bone specific alkaline phosphatase (BALP), and bone gla protein(BGP) at the time of killing. The femora were taken for three-point bending test, measuring maximal loading, flexibility loading, flexibility strain, and maximal strain. Results (1) Four weeks after the administration, the level of BALP in the OVX-FSIM group was higher than that in the OVX group (P〈0.05) and its flexibility loading was lower than that of the OVX group (P〈0.05) ; (2) 12 weeks after the administration, the level of BLAP and BGP in the OVX+ SIM group was higher than that of the OVX group(P〈0.01) and its maximal loading and flexibility load- ing were higher those of the OVX group (P〈0.05 or P〈0. 01); (3) the comparison of the two time points showed that, 12 weeks after the administration, the level of BALP in the OVX-i-SIM group was significantly decreased (P〈0.01) but the level of BGP, the maximal loading, and the flexibility loading of the OVX-FSIM group were significantly increased(P〈0.01). Conclusion Simvastain can affect bone metabolism, increase bone formation and improve biomechanical characteristics of femora of rats with ovariectomy-induced osteoporosis
