摘 要:目的:观察解毒化瘀方对内毒素血症(endotoxemia,ETM)大鼠肝细胞线粒体COX的影响。方法:将56只大鼠随机分为正常对照组及内毒素(脂多糖LPS注射后)6小时、12小时、24小时组和解毒化瘀方6小时、12小时、24小时组。取肝组织分离线粒体,采用紫外分光光度计检测COX活性,采用RT-PCR法检测COXⅠ、COXⅡ、COXⅢ的表达变化。结果:内毒素造模后6小时、12小时、24小时,肝线粒体内COX活性明显下降,与正常组相比有统计学意义(P<0.01),24小时降为最低。解毒化瘀方组与内毒素组相同时相点比较,解毒化瘀方12小时组、24小时组COX活性增加,两组比较差异有显著性意义(P<0.05)。内毒素组6小时、12小时、24小时,COXⅠmRNA、COXⅡmRNA表达逐步下降,与正常组相比有统计学意义(P<0.01),COXⅢmRNA表达与正常组比较无统计学意义(P>0.05)。解毒化瘀方24小时组COXⅠmRNA表达高于内毒素同时相组(P<0.05)。结论:解毒化瘀方能提高COX活性,上调线粒体编码基因COX表达水平,这可能是改善能量代谢、保护肝细胞的一个作用机制。Objective: To observe the effect of Jiedu Huayu Fang on hepatocyte mitochondrial COX in endotoxemia rats. Methods: Fifty six healthy adult rats were randomly divided into normal control group and 6, 12, 24 hours groups after injecting LPS and Jiedu Huayu Fang treatment group, separated hepatocyts mitochondria, then examined the activity of COX in the mitochondria of liver cell and examined the changes of the expression of liver mitochondria mtDNA COX Ⅰ , COX Ⅱ , COX Ⅲ mRNA through the method of RT-PCR. Results: After endotoxin, the activity of COX descended obviously in 6h, 12h, 24h groups, comparing with the normal group, which had statistics meaning ( P 〈0. 01 ) . The activity of COX in 12h, 24h groups of Jiedu Huayu Fang, comparing with the groups of corresponding endotoxin, were better increased than in the groups of corresponding endotoxin (P 〈0. 01 or P 〈0. 05) . The expression of mitochondria COX Ⅰ mRNA in 6h, 12h, 24h groups descended gradually, comparing with the normal group, which had statistics meaning (P 〈 0.01 ) , the expression of mitochondria COX Ⅱ mRNA descended gradually too ( (P 〈 0. 01 ) . Comparing with the normal group, the expression of mitoehondria COX Ⅲ mRNA had not statistics meaning ( P 〈 0. 05 ) . The expession of COX Ⅰ mRNA in 24h groups of Jiedu Huayu Fang, comparing with the groups of corresponding endotoxin, was more increased ( P 〈 0. 05 ) . Conclusion: Jiedu Huayu Fang can significantly raise the activity of COX and the expression of COX on hepatocyte mitochondrial in endotoxemia rats, which probably ameliorate mitochondrial energy metabolism obstacle, having the function of protect the liver cell.
