湖北汉族人群15个STR基因座的遗传多态性

摘　　要：目的调查湖北汉族人群15个STR基因座(D8S1179、D21S11、D7S820、CSF1PO、D3S1358、TH01、D13S317、D16S539、D2S1338、D19S433、vWA,TPOX,D18S51,D5S818,FGA)的遗传多态性分布。方法采用Identifiler试剂盒复合扩增305例湖北汉族个体的15个STR基因座,产物经3130遗传分析仪电泳分离和分型检测,分析检验基因座的遗传学参数:基因频率、杂合度、多态信息含量(PIC)、个体识别能力(DP)、非父排除概率(PE)和遗传平衡状态。结果305名无关个体共检测到15个STR基因座的146个等位基因,各基因座基因型分布均符合Hardy-Weinberg平衡(P>0.05)。13个STR基因座的PIC>0.5,14个STR基因座的DP>0.8,12个STR基因座的PE>0.5;累积个体识别力>0.999 999 999,累积非父排除率>0.999 998 8。结论湖北汉族人群15个STR基因座的绝大多数基因座具有较高的遗传多态性,其群体遗传学数据可用于本地人类遗传学、法医亲子鉴定和个体识别等研究领域。Objective To study the genetic polymorphisms of 15 short tandem repeat （STR）loci （D8S1179, D21Sll, D7S820, CSF1PO, D3S1358, TH01, D13S317, D16S539, D2S1338, D19S433, vWA, TPOX, D18S51, D5 $818, FGA） in Han population of Hubei province. Methods The genotypes of 15 STR loci in Han population of Hubei province were generated by mul tiplexing PCR using the Identifier PCR system and genotyping using 3130 DNA genetic sequencer. Allele frequency, heterozygosity（ H ）, polymorphic information content（PIC）, discrimination power（ DP）, probability of exclusion（ PE）, cumulative proba bility of exclusion（CPE）and cumulative discrimination power（CDP）were calculated. The genetic balance of 15 STR loci was tested. Results Out of a total of 305 individuals, 146 alleles（15 STR loci）were detected. The genotype distribution of the 15 STR loci fitted Hardy--Weinberg balance（P〉0.05）. PIC of 13 STR loci 〉0.5,DP of 14 STR loci 〉0.8,PE of 12 STR loci 0.5,CDP〉0. 999 999 999,and CPE〉0. 999 998 8. Conclusion Most of 15 STR loci in Han population of Hubei province have higher genetic polymorphisms,and can he used as the genetic markers of Han population in the studies of population genetics, individual identification and paternity test in forensic science.
