摘 要:目的:建立HPLC法,对庚铂R-和S-异构体进行拆分并检测其中S-异构体含量。方法:色谱柱采用Chiro-biotic R手性柱(Astec,250 mm×4.6 mm,5μm);流动相为0.1%三乙胺溶液(用乙酸调节pH为6.8)-异丙醇(55∶45);流速为0.5 mL.min-1;柱温为30℃;检测波长210 nm;进样量10μL。结果:本法对庚铂异构体的分离度为2.1,S-异构体的线性范围为0.51~10.20 mg.L-1(r=0.999 8),检测限为0.2 ng,平均回收率为99.1%(RSD=0.65%,n=9)。结论:该方法操作简便,分离效果好,可用于庚铂中S-异构体含量测定。Objective: To establish an HPLC method for separation of the R-and S-heptaplatin and determination of the S-isomer.Methods: The separation was performed on a Chirobiotic R chiral column(Astec,250 mm×4.6 mm,5 μm).0.1% Triethylamine solution(adjusted to pH 6.8 with acetic acid)-iso-propylalcohol(55∶45) was used as mobile phase.The flow rate was 0.5 mL ·min-1.The column temperature was 30 ℃.The detection wavelength was set at 210 nm.The injection volume was 10 μL.Results: This method showed that the resolution of the isomer was 2.1,the linear range of the S-isomer was 0.51~10.20 mg ·L-1(r =0.9998),the detection limit was 0.2 ng and the average recovery rate was 99.1%(RSD=0.65%,n=9).Conclusion: The method is simple and effective.It can be used for content determination of S-isomer in heptaplatin.
