摘 要:目的:观察四妙勇安汤是否对动脉粥样硬化(AS)家兔模型核因子-κB(NF-κB)表达及相关因子有调控作用,从而拮抗AS的发生和斑块的形成。方法:56只雄性日本大耳白家兔随机分成空白对照组、模型组、辛伐他汀组和四妙勇安汤组,除空白对照组外,其余各组建立主动脉粥样硬化斑块模型,从实验第1d开始予相应药物干预,第10周末麻醉处死后取主动脉行NF-κB免疫组化染色观察,并于实验0,3,6,10周动态检测血清中TNF-α、IL-1及MCP-1含量。结果:模型组粥样斑块中泡沫细胞NF-κBp65亚基表达丰富,用药组NF-κBp65亚基较模型组阳性细胞表达减少,四妙勇安汤对斑块区NF-κBp65亚基的表达有明显的抑制。模型组各时间点血清中TNF-α、IL-1和MCP-1含量显著增高(P<0.01),两给药组在各时间点均有不同程度的降低IL-1、TNF-α和MCP-1水平的作用(P<0.05或P<0.01)。结论:四妙勇安汤可减少斑块区NF-κBp65亚基表达,同时具有减少炎症因子释放的作用。Objective: To observe whether the Simiao Yongan Decoction regulate the nuclear factor-κB (NF-κB) expression and related factors in atherosclerosis (AS) rabbit model, thus restrain the occurrence of atherosclerosis and plaque formation. Methods: 56 male Japanese rabbits were randomly divided into normal group, model group, Simvastatin group and Simiao Yongan Decoction group, in addition to the normal group, other groups were established aortic atherosclerotic plaque model. From thelst day of experiment, the corresponding drug intervention began, 10 weeks later, the rabbits were killed over the weekend after the anesthesia line access aortic NF-κB immunohistochemical staining, and the experimental dynamic 0,3,6,10 weeks in serum TNF-α, IL-1 and MCP-1 content. Results: The expression of NF-κB p65 subunit in model group was rich, and compared with it, the expression of NF-κB p65 subunit in treatment groups decreased, and the Simiao Yongan Decoction can obviously inhibit the expression of NF-κB p65 subunit. In model group, the expressions of IL-1, TNF-α and MCP -1 in serum increased significantly at different time points (P〈0.01). In two treatment groups, the expression of IL-1, TNF-α and MCP -1 at different time points decreased at different extents (P〈0.05 or P〈0.01). Conclusion: Simiao Yongan Decoction can reduce NF-κB p65 subunit expression on plaque area, and reduce the release of inflammatory factors.
