摘 要:目的:测定葛根黄豆苷元在不同pH值下的平衡溶解度和油水分配系数(P)。方法:采用饱和法及摇瓶法测定表观溶解度,通过药物分配平衡后在油相和水相中的浓度比,计算油水分配系数。结果:葛根黄豆苷元在不同pH条件下的平衡溶解度(S)分别为:4.705、3.451、4.792、4.248、6.118、15.291、23.450mg/L,在不同pH条件下的油水分配系数分别:1.339、1.323、1.081、1.116、1.515、0.604、1.709。结论:药物在水中的平衡溶解度及油水分配系数与介质的pH值有关,pH>6.8时平衡溶解度急剧上升,当6.8<pH<7.8时,药物在油相中分配较多。Objective: To determine the equilibrium solubility and oil/water apparent partition coefficient (P) of daidzein in a series of test solutions of different pH. Methods: The apparent solubility was determined by saturation method, and P was calculated with concentration ratio of drag in oil and water phase after partition equilibrium. Results: The equilibrium solubility of daidzein in different pH respectively was 4.705, 3.451, 4.792, 4.248, 6.118, 15.291, 23.450 rag/L, and the oil/water apparent partition coefficient (P)of daidzein in different pH respectively was 1.339, 1.323, 1.081, 1.116, 1.515, 0.604, 1.709. Conclusion: The equilibrium solubility and apparent partition coefficient of the drug were correlated to the pH of medium. The equilibrium solubility of daidzein sharply rose up when pH〉6.8, and the drug was more distributed in the lipid phase when 6.8〈pH〈7.8.
