摘  要:目的:前期的研究已证实中药掌叶半夏提取物的有效部位(pinellia extract fraction,PE)对体外培养的宫颈癌细胞株CaSki和HeLa有促凋亡作用,通过本次研究进一步明确PE对CaSki的促凋亡作用机制。方法:采用RT-PCR方法检测0.15g/L的PE作用24h后CaSki细胞Bcl-2和Bax mRNA表达的变化,采用Western Blot检测0.15g/LPE作用24h后CaSki细胞Bcl-2、Bax蛋白表达的变化。结果:0.15g/L的PE作用24h后CaSki细胞Bcl-2 mRNA和蛋白的表达均明显下降,而Bax mRNA和蛋白的表达均明显上升。结论:PE对CaSki宫颈癌细胞株促凋亡作用之一是抑制Bcl-2基因的表达、促进Bax基因的表达。这为进一步研究PE的抗宫颈癌作用机制以及新药的研制开发奠定了一定的实验基础。Objective: To investigate the mechanisms of apoptosis induction effect of Pinellia extract fraction (PE) on CaSki cell line of cervical cancer cultured in vitro, based on the previous result of PE's effect on CaSki and HeLa. Methods: Expression of Bcl-2 and Bax mRNA were detected by RT-PCR; Expressions of Bcl-2 and Bax protein were detected by western blot assay Results: Expressions of Bcl-2 mRNA and protein were down-regulated, while expressions of Bax mRNA and protein were up-regulated after treated with 0.15g/L PE for 24 hours. Conclusion: PE can induce apoptosis of CaSki cells. One of the mechanisms was by down-regulate expression of Bcl-2 and up-regulate expression of Bax. This result will provide a certain experiment basis for the research of PE's antitumor effects.
