定期无偿献血者T细胞免疫功能的研究

摘　　要：目的通过对定期无偿全血捐献者、定期无偿血小板捐献者2个特殊健康人群的T细胞数量以及细胞活化、细胞因子分泌功能的研究,评价定期献血对机体T细胞免疫功能的影响。方法采用流式细胞术对3组人群(定期无偿全血捐献者、定期无偿血小板捐献者、对照组)外周血中T细胞总数、CD4+T细胞群及CD8+T细胞群进行数量分析;分离外周血淋巴细胞体外培养,加丝裂原植物凝集素PHA刺激T细胞活化,流式细胞术检测CD4+CD25+活化T细胞群;Lum inex 100多功能液相分析平台检测细胞因子IL-2的分泌量。结果定期无偿全血捐献组及定期无偿血小板捐献组的T细胞总数及CD4+T细胞群、CD8+T细胞群的数量与对照组的差异无统计学意义(P>0.05);T细胞活化指标CD25+、CD69+均在培养d4(72h)表达最高;定期无偿捐献全血组CD4+CD25+活化T细胞以及IL-2分泌量与对照组的差异无统计学意义(P>0.05);定期无偿捐献血小板组CD4+CD25+活化T细胞以及IL-2的分泌量高于对照组(P<0.05)。结论机体通过强大的正负免疫调节,使定期献血者T细胞的数量和功能维持稳定状态。Objective Through research on T cell numbers and cell activation, cytokine secretion of regular whole blood volunteers and regular platelet volunteers, we evaluate the impact of regular donating blood as a stimulating factor to the body＇s T cells immune function. Methods The numbers of T cells, CD4＋ T cells and CD8 ＋ T cells in peripheral blood of three groups of people（regular whole blood volunteers, regular platelet volunteers, control group）were counted by flow cytometer （FCM）. Peripheral blood lymphocytes were separated and cultured in vitro, then phytohemagglutinin （PHA） added to stimulate the activation of original T-cells, CD4＋ CD25 ＋ Activated T cells groups were counted by FCM. Secretion of IL-2 was detected by Luminex 100, a muhifunctional platform. Results There was no significant statistical difference of in the total number of T cells and, the number of the CD4＋ T cells and CD8＋ Tcells between the group of free volunteer regular whole blood donations, the group of free to donate volunteer platelet donation as compared to the control group （P 〉 0. 05）. T cell activation expresses showed the highest in the fourth day （72 h） of culture. There was no significant difference of in CD4＋ CD25 ＋ activated T cells numbers, and IL-2 secretion level between a regular basis to donate volunteer whole blood donation group and the control group（P 〉0.05）. The CD4＋ CD25 ＋ activated T cells number and IL-2 secretion level of regular volunteer platelet donation group crowd was significantly higher than the control group＇s （P 〈 0. 05）. Conclusion Intensely positive and negative immune regulation keeps the number and function of T cell of regular blood donors in balance.
