参桂胶囊对急性心肌缺血大鼠心肌蛋白质组学的影响

摘　　要：目的:观察参桂胶囊对舌下静脉注射垂体后叶素(pituitrin,PIT)诱发大鼠心肌缺血的心电图及心肌蛋白质组谱的影响。方法:采用大鼠舌下静脉注射垂体后叶素造成急性心肌缺血模型,观察各组注射垂体后叶素后不同时间点第Ⅱ导联心电图T波变化;采用双向凝胶电泳(two-dimensional protein electrophoresis,2DE)及计算机图像分析系统对心肌总蛋白进行分离分析。结果:参桂胶囊能够明显抑制垂体后叶素引起的心电图T波的改变,各组间蛋白质斑点发生显著性变化,主要表现为蛋白点的缺失、增加或者表达量的改变。结论:参桂胶囊能明显改善垂体后叶素引起的大鼠心肌缺血症状;缺血后,大鼠心肌蛋白表达谱发生改变,发生改变的蛋白质可能与心肌组织的损伤与保护有关,而参桂胶囊可使这种改变减轻。Objective：to study the effect of Shengui Capsule on acute myocardial ischemia induced by pituitrin in rats and Myocardial proteome Spectrum.Methods：Pituitary（0.35 U/kg） was given to model of acute myocardium ischemia.The changes of theⅡroad electrocardiogram（T wave）;The use of two-dimensional gel electrophoresis and computer image analysis system for Myocardial of total protein were separated.Results：Copy the model of myocardial ischemia successfully,Shengui Capsule can inhibit pituitrin-induced T-wave changes in ECG,protein spots among the three groups significant changes occurred,the main point of realization for the protein lack of an increase or change in expression.Conclusion：Shengui Capsule can improve pituitrin-induced myocardial ischemia symptoms;myocardial ischemia,the animals of myocardial protein expression profile changes,changes in the protein may be associated with myocardial injury and protection,and Shengui Capsule can mitigate this change.
