摘 要:目的探讨瘦素、纤连蛋白对早孕绒毛滋养细胞分泌基质金属蛋白酶的影响及其作用。方法从孕6-8周的人工流产胚胎绒毛中分离、纯化出细胞滋养细胞(CTB)体外培养,在transwell上室CTB培养液中分别加入不同浓度lep-tin,在transwell下室中加入50μg/mlFN,采用明胶酶谱分析法检测培养液中CTB分泌的MMP-2、MMP-9值并进行比较。结果FN组、协同组分泌的MMP-2,9显著高于对照组(P<0.05),协同组又高于单加leptin或FN组(P<0.05)。结论leptin、FN均具有促进CTB分泌MMPs的作用,且leptin与FN协同作用,更加强了CTB对MMPs的分泌,从而参予调控CTB的浸润能力。Objective:To explore the effects of Leptin and Fibronectin on the secretion of Matrix metalloproteinase in trophoblsts in human first trimester gestation.Methods:Isolated and sublimated Cytotrophoblasts(CTB) from chorionic villi at 6-8 weeks by abortion were cultured in vitro. The CTB cultured in the upper chambers of transwell were added different concentration of leptin,and 50μg/ml FN were added in the lower chamber. MMP-2 and MMP-9 secreted by CTB was detected by Gelatinase spectrum analysis. Results:The purity quotient ANOVA showed significant difference of MMP-2 and MMP-9 levels in groups,P〈0.05.Conclusions:Leptin and FN synergisticly action on CTB could influence the secretion of Matrix metalloproteinases,and so to regulate the invasive ability of CTB.
