摘  要:目的探讨核因子-кB(Nucler factorкB,NF-кB)和TNF-a在鼻息肉中的表达及意义。方法取32例鼻息肉患者的息肉组织,并以其中的14例患者的鼻中隔黏膜作对照,用凝胶迁移实验(Electro-phoretic mobility shift assay,EMSA)检测核因子кB的DNA结合活性,用RT-PCR技术检测TNF-a mRNA的表达,对NF-кB的DNA结合活性与TNF-a mRNA行Pearson相关性分析。结果鼻息肉组织中NF-кB的DNA结合活性升高,明显高于对照组(P<0.01);鼻息肉组织中TNF-a mRNA的转录明显高于对照组(P<0.01);Pearson相关性分析提示,鼻息肉组织中NF-κB的DNA结合活性与TNF-a mRNA的转录呈显著正相关(r=0.793,P<0.01)。结论鼻息肉组织中NF-кB的激活上调了TNF-a mRNA的转录,NF-кB和TNF-a可能是导致鼻息肉局部微环境变化的重要因素。Objective To evaluate the expression of nuclear factor - κB and tumor necrosis factor - a in the nasal polyps tissue and their significance. Methods The specimens from 32 nasal polyps and 14 nasal septum mucous membranes were studied by electrophoretic mobility shift assay(EMSA) for the activity of NF - κB, and by reverse - transcription polymerase chain reaction ( RT - PCR) for the expression of TNF - a mRNA. And the correla- tions between the activity of NF - κB and TNF - a mRNA were evaluated by Pearson correlation analysis. Results The activity of NF - κB DNA were significantly increased in nasal polyps than in nasal septum ( P 〈 0.01 ), and the expression of TNF - a mRNA were significantly increased in nasal polyps than in controls ( P 〈 0.01 ). The Pearson correlation analysis demonstrated that there were positive relationship between the activity of NF - κB DNA and the expression of TNF - a mRNA ( r = 0. 793, P 〈 0.01 ). Conclusion These results suggest that the activity of NF - κB in nasal polyps may up - regulated the transcription level of TNF - a mRNA significantly. NF - κB and TNF - a may play important roles in the changes of microenvironment of nasal polyps.
