摘 要:目的:探讨天麻遗传变异与其品质的关系,为优良种质的选育提供科学途径。方法:采用CTAB法提取天麻DNA,进行PCR扩增,用微卫星DNA分子技术(SSR)标记DNA;用高效液相色谱法检测不同种质天麻的天麻素含量,分析两者之间的关系。结果:出现A条带类型的样品天麻素含量较高,7EKYC、9EDF;24GBJ、22GZJ;1FDJ、2FLL;4VDF、6EKYW;18GWAC、8EWD,分别聚到一起,遗传距离较小。结论:特异引物扩增位点与天麻素含量有关,高天麻素含量与SSR高分子量条带有关,其关系有待进一步研究。Objective: To investigate the associativity between the genetics and variation of Gastrodia elata Blume and its quality, so as to provide scientific path for good species selection. Methods: DNA of Gastrodia elata Blume was extracted by CTAB method, amplified by PCR and marked by SSR technique; Gastrodin content was detected by HPLC. The relation between SSR fingerprinting and Gastrodin contents was analyzed. Results: Gastrodin contents were relatively high in specimens with type A bands. Genetic distances between 7EKYC and 9EDF, 24GBJ and 22GZJ, 1FDJ and 2FLL, 4VDF and 6EKYW, 18GWAC and 8EWD were small. Conclusions: The sites for special primer amplification relate to Gastrodin content, and Gastrodin content relates to SSR fingerprinting, which needs further research.
