摘  要:目的:研究黄芪多糖的抗肿瘤活性及对荷瘤小鼠血清中IL-2,TNF-α水平的影响。方法:以人肿瘤细胞株为模型,采用cell counting kit-8(CCK-8)检测体外抗肿瘤活性;以H22和S180为模型,检测体内抗肿瘤活性;ELISA法检测对荷瘤小鼠血清中IL-2和TNF-α水平的影响。结果:黄芪多糖对K562,SMMC-7721,HCT116和A549细胞的增殖无明显影响,在50,100 mg.kg-1时,对H22的抑瘤率分别为32.84%,45.09%,对S180的抑瘤率分别为36.55%,50.35%,且能提高荷瘤小鼠血清中IL-2和TNF-α的水平。结论:黄芪多糖无明显细胞毒活性,但体内抗肿瘤活性较强,其机制与增强机体的免疫功能有关。Objective: To study the antitumor activity of Astragalus Polysaccharides and the levels of IL-2 and TNF-α in tumor-bearing mice. Methods:The cytotoxicity on human tumor cell lines was determined by cell counting kit-8 (CCK-8) assay. The inhibitory effect on tumor growth was observed by the models of hepatoma 22 (H22) and implanted sarcoma 180 (S180) in mice. The levels of IL-2 and TNF-α in tumor-bearing mice were investigated by ELISA assay. Results:Astragalus Polysaccharides had no effect on the proliferation of K562, SMMC- 7721, HCT116 and A549 cells. At doses of 50 and 100 mg· kg ^-1 ,the inhibitory rates of Astragalus Polysaccharides on H22 and S180 were 32.84% , 45.09% and 36.55% , 50.35% , respectively. Astragalus Polysaccharides enhanced the expression levels of IL-2 and TNF-α in tumor-bearing mice. Conclusion: Astragalus Polysaccharides had no cytotoxicity, but exhibited anti-tumor effect in vivo, and the mechanism may be related to enhancing the immune function.
