Metabolism of Polyamines During the Development of Panicles in Cytoplasmic Male-Sterile Line and Its Maintainer Linein Oryza sativa L.

摘  要:在幼穗发育过程中,不育系和保持系幼穗多胺含量先剧降后稳定或略回升,精氨酸脱羧酶活性快速下降,而二胶和多胺氧化酶活性缓慢下降。从雌雄蕊形成期到花粉母细胞形成期,不育系的多胺含量和精氨酸脱羧酶活性明显低于保持系;不过,两系二胺氧化酶和多胺氧化酶活性却差别不大。外施D-Arg抑制两系Put合成,也抑制以Put为前体的Spd的合成;外施MGBG抑制Spd和Spm的合成;同时,D-Arg或MGBG对不育系花粉育性影响不大,但明显降低保持系花粉育性,D-Arg+MGBG对花粉育性的降低效应更强;Put和pd+Spm可抵消(或部分抵消)D-Arg和MGBG的降低效应。且Put+Spd+Spm能使不育系花粉的育性得以轻度恢复。By using HPLC, Changes of polyamines contents during the devlopment of panicle in cytoplasmic male -sterile line and its maintainer line in rice were studied. The activities of ADC,key enzymes in the biosythsis of polyamines in plants, and DAO, an enzyem is related to the degradation of Put,and PAO, an enzym involved in the catabolism of Spd and Spm, were also studied. The effects of D-Arg an inhibitor of Put formation , and MGBG, an inhibitor of the biosynthesis of Spd and Spm, on the contents of polyamine in panicles and fertility of pollen in both lines were studied, too. The results were as follow: From the stage of stamen and pistil formation to the stage of pollen mother cell meiosis, contents of all 3 polyamins measared in the panicles of both lines decreased quickly , followed by a slightly increase in content of Put and Spd. However, from the formation stage of stamn and pishl to that of polln mother cell, level of polyamine in the maintainer line was much higher than tha in CMS line (Fig. 1). The activity of ADC decreased from formation stage of stamen and pistil to filling stage of pollen in both 2 lines, and it was much higher in the maintainer line than in CMS line,too(Fig. 2). The activities of DAO and PAO in both 2 lines decreased during the whole course, and these in CMS line were almost the same as in its maintainer line(Fig. 3). It is indicated tha the difference in polyamines content between CMS line and its maintainer line could result from the difference in activity of ADC. D-Arg reduced Put and Spd contents, and MGBG decreased Spd and Spm levels and increased Put level(Table 1). And these 2 inhinbit0rs cut down the fenility of pollen in maintainer line markedly (Fig- 4). Put or Spd +Spm reduced the inhibiting effect of DArg or MGBG on PAs biosynthesis and restored the fertility of pollen (Fig. 4).Put + Spd + Spm also restored the fertility of pollen in CMS line slighily(Fig. 4) It is suggested that insufficiency of polyamines in the panicles of CMS line might be related to
