摘  要:血清中元素不同形态含量变化与人体营养和健康有着密切关系。文章利用两步沉淀蛋白质-石墨炉原子吸收法测定了血清中非蛋白结合的铜和锌。无水乙醇以2∶1与血清混合后,再经70℃温度变性处理,通过贝克曼离心机高速离心,可将蛋白质完全沉降。在血清中加入EDTA可将白蛋白结合的铜和锌脱落,转化为非蛋白结合铜和锌,从而可测得球蛋白和白蛋白分别结合的铜和锌含量。用氘灯校正背景,石墨炉原子吸收法进行测定,方法检测限:Cu,1.2μg.L-1 Zn,0.098μg.L-1。回收率:Cu,92.3%~104%,Zn,90%~107%。利用此方法测定了健康人及肿瘤患者血清和肝豆状核性患者中球蛋白、白蛋白及结合的铜与锌以及游离的铜和锌,并与健康人作了对比。A simple and reliable method was developed for the determination of serum copper and zinc in different chemical forms by graphite furnace atomic absorption spectrometry (GFAAS) with ethanol-EDTA precipitation. The serum and ethanol were mixed with volume ratio 1 : 2. The mixture was incubated at 70 ℃ and ultra-centrifuged to precipitate proteins. Zinc and copper can be released from albumin after EDTA treating. Thus, macroglobulin-zinc, ceruloplasmin-copper, and albumin-bound zinc or copper could be determined by two proposed precipitation steps. The determination limit of copper (3a) was 1.2μg · L^-1 and the recovery was 92.3%-104%, while the determination limit of zinc (3a) was 0. 098 μg · L^-1, and the recovery was 90%- 107%. This two-step precipitation method can be used to determine serum zinc and copper in various chemical forms in tumor, Wilson patients and heathy human.
