旋覆代赭汤含药血清对胃窦平滑肌细胞IP_3受体及钙离子浓度影响的实验研究

摘　　要：目的:观察旋覆代赭汤含药血清对大鼠胃窦平滑肌细胞(SMC)IP3受体(IP3R)及钙离子(Ca2+)浓度的影响。方法:分离大鼠胃窦SMC,加入IP3受体拮抗剂肝素后,镜下观察含药血清对胃窦SMC收缩作用;检测细胞Ca2+总浓度。结果:IP3受体拮抗剂肝素可抑制旋覆代赭汤含药血清对胃窦SMC的收缩作用;各剂量组与无钙缓冲液组的细胞内钙离子总浓度相比较无明显差异。结论:旋覆代赭汤含药血清可使胃窦SMC收缩,其机制可能是通过IP3通路作用于细胞,引起细胞内质网释放Ca2+使胃窦SMCCa2+升高。Objective： To observe the effect on XuanFu DaiZhe （XFDZ） decoction containing serum of Gastric antral smooth muscle cells （SMC） IP3 receptors （IP3R） and concentration of calcium ion （Ca^2＋）. Methods： The gastric antrum SMC of rats was isolated, the heparin of IP3R antagonist was added, the contraction of gastric antrum SMC with serum containing medicine was observed by the microscopic, and the total concentration of cellular Ca^2＋ was detected. Results： The heparin of IP3R antagonist can inhibit the contraction of gastric antral SMC with XFDZ decoction containing serum. There are no obvious difference in the total concentration of intracellular calcium between every dose group of XFDZ decoction and calcium-flee buffer group. Conclusion： XFDZ decoction can make gastric antral SMC contract. Its mechanism may be related to the increasing gastric antral SMC Ca^2＋ because endoplasmic reticulum releases Ca^2＋ by IP3 pathways.
