分化抗原6A8在B淋巴细胞膜和胞浆中的表达

摘　　要：采用间接免疫荧光染色、流式细胞仪、激光扫描共聚焦显微镜、Ｎｏｒｔｈｅｒｎｂｌｏｔ及Ｗｅｓｔｅｒｎｂｌｏｔ分析研究分化抗原６Ａ８在人淋巴细胞的表达。流式细胞仪分析显示，单抗６Ａ８与所检测的Ｂ细胞株３Ｄ５、ＣＥＳＳ和Ｄａｕｄｉ有反应，与人组织细胞瘤细胞株Ｕ９３７也有反应，与慢性髓性白血病细胞株Ｋ５６２及所检测的Ｔ细胞株Ｊｕｒｋａｔ和Ｐｅｅｒ不反应。Ｎｏｒｔｈｅｒｎｂｌｏｔ检测支持上述现象，３Ｄ５和Ｕ９３７细胞ｍＲＮＡ转录较丰富，ＣＥＳＳ细胞次之，而Ｊｕｒｋａｔ与Ｐｅｅｒ细胞的ｍＲＮＡ转录很少。激光扫描共聚焦显微镜观察显示，单抗６Ａ８识别的分化抗原见于３Ｄ５细胞浆和胞膜。可见在人Ｂ细胞株３Ｄ５，分化抗原６Ａ８不仅表达于胞膜，也表达于胞浆。Study on the expression of differentiation antigen 6A8 on the membrane and in the cytoplasm of B lymphocytes. Observation of indirect fluorescence staining upon a flowcytometer and a lazer scan confocal microscope, Northern blot and western blot. Flowcytometric analysis demonstrated that B cell line 3D5, CESS, Daudi and a histocytoma cell line U937 were positively stained, K562 cell and T cell line Jurkat. Peer were negatively stanined by McAb 6A8. Northern blot confirmed the discovery in that the mRNA expressed in 3D5 cell, CESS cell and U937 cell is rich, and in Jurkat and Peer cell is poor. The examination upon a lazer scan confocal microscope revealed the expression of the differentiation antigen recognized by McAb 6A8 both on the membrane and in the cytoplasm of 3D5 cell. Differentiation antigen 6A8 is expressed both on the membrane and in the cytoplasm of 3D5 cell.
