与人精子有共同抗原的临床细菌分离株的筛选

摘　　要：以ＲＡＨＳ－Ａｂ与４９３株临床细菌分离株进行玻片凝集反应，筛选出４种１１株可与ＲＡＨＳ－Ａｂ发生明显凝集反应的细菌株。选择其中Ｅ．ｃｏｌｉ５０６株作进一步研究。制备Ｅ．ｃｏｌｉ５０６株全菌蛋白进行包被，ＥＬＩＳＡ检测可与ＲＡＨＳ－Ａｂ出现阳性反应。进一步将全菌蛋白与ＲＡＨＳ－Ａｂ进行Ｗｅｓｔｅｒｎ印迹反应，发现可出现数处阳性反应条带，其中位于３６ｋＤ和６７ｋＤ的两个条带反应最为明显。制备人精子蛋白并包被，以Ｅ．ｃｏｌｉ５０６株全菌抗体进行ＥＬＩＳＡ检测，发现二者可以发生免疫反应。这些结果证实某些细菌如Ｅ．ｃｏｌｉ５０６株与人精子间存在共同抗原。The rabbit anti human sperm antibody(RAHS Ab) was prepared to react with 493 strains of clinical bacterial isolates by slide agglutination test.Eleven positive strains were screened.One of the positive strains,E. coli 506,was selected to go on further study.The whole cell proteins of E. coli 506 prepared and precoated on the surface of microtiter plate.It was found that the proteins could react with RAHS Ab by indirect ELISA.The whole cell proteins of E. coli 506 strain were electrophoretically transferred to the nitrocellulose membrane from the SDS PAGE gel using the MiniTrans Blot cell.Several bands of the proteins were found to react with RAHS Ab by Western blotting.Two of the bands,which located in 36 kD and 67 kD respectively,were found to react with RAHS Ab strongly.On the other hand,the whole cell proteins of human sperm were found to react with anti E. coli 506 strain antibody by ELISA.The results show that some bacterial strains,such as E. coli 506 strain,may have common antigens with human sperm.
