摘 要:为了阐明己克隆的水稻C端结合蛋白家族基因O sC tBP-A的功能,构建了该基因的过量表达和RNA i沉默载体,通过农杆菌介导转化水稻品种日本晴愈伤组织,分别获得了45个过量表达和75个RNA i沉默T0代转基因植株,并进行了PCR和GUS染色鉴定。T1代过量表达株O sC tBP-A基因表达量明显增加,苗期根系生长增加,对水稻白叶枯病抗性无明显变化;而基因沉默株O sC tBP-A基因表达量显著下降,苗期根系发育迟缓,抗旱性明显降低,抗病性减弱。表明O sC tBP-A基因的表达可能对水稻苗期根系发育、抗旱性和抗病性具有不同程度的调控作用。In order to expound the functions of cloned C-terminal binding protein gene OsCtBP-A,its overexpression and RNAi constructs were constructed and transformed into calli of rice(cv.Nipponbare) via Agrobacterium-mediated transformation.Forty-five transgenic plants with overexpression and 75 with RNAi were generated and identified by PCR and GUS staining analysis.In T1 progeny of transgenic rice lines with overexpression,OsCtBP-A gene expression was remarkably increased,and root development was enhanced during seedling stage,while its resistance to rice leaf blight was no much changed.In contrast,OsCtBP-A gene expression was significantly reduced,and the root development at seedling stage was slowed down and its resistance to drought was weakened in RNAi silenced lines.The results suggested that OsCtBP-A gene might be involved in regulation of root development at seedling stage and resistances to drought and diseases.
