摘 要:目的研究骨髓间充质干细胞(MSCs)移植对大鼠脊髓损伤的作用。方法将大鼠MSCs移植到大鼠脊髓损伤处,移植后评估大鼠行为学改变,免疫组织化学染色法观察BrdU标记的MSCs在脊髓损伤处存活情况,辣根过氧化物酶(HRP)逆行示踪,观察脊髓损伤处神经通路的重构建,皮层体感诱发电位(CSEP)测定脊髓损伤处神经通路传导功能的恢复。结果术后移植组评分均高于损伤组,移植组大鼠脊髓损伤区远端检测到BrdU阳性细胞,脊髓损伤近侧端检测到HRP阳性细胞,CSEP波形恢复但波幅减低,潜伏时延长。结论MSCs可在损伤区存活并分化为神经元,在损伤局部形成神经元通路,使损伤脊髓的远端和近端建立联系,神经纤维传导功能部分恢复。Objective To study the effects of bone marrow mesenchymal stem cells transplantation on functional recovery of the injured rats spinal cord. Methods MSCs labeled with Brdu were transplanted into rats model of spinal cord half-transection injury. The open-field BBB scoring system was employed to evaluate behavioral changes. MSCs’survival after transplantation was identified by BrdU immunohistochemistry. We observed the reconstruction of neuronal circuits by HRP coloration. The recovery of transduction function after spinal injury was examined by cortex somatosensory evoked potential (CSEP). Results Treated rats generally showed better functional recovery than control rats after operation. BrdU-positive cells could be found in the spinal cord injury site one week after transplantation. At two months after transplantation,HRP-positive cells could be found at rostral of the spinal cord injury site of treated rats,but not be found in control. CSEP could be evoked at treated rats from two months after transplantation,but not in controls. Conclusion MSCs may survive in the spinal cord injury site via local injection immediately after spinal cord injury,and may promote regeneration of the injured axons.
