摘 要:目的探讨煤焦油对小鼠肺成纤维细胞(NI H/3T3)的细胞间隙连接通讯(GJIC)功能的影响。方法以小鼠NI H/3T3细胞为靶细胞,煤焦油对NI H/3T3细胞GJIC的影响采用划痕标记染料示踪(SLDT)技术测定。结果煤焦油低、中、高剂量(0.1、1.5、22.5μg/ml)作用6 h后,高剂量组与阴性对照组比较,GJIC的平均水平下降,差异有统计学意义(P<0.05);随着作用时间延长至12 h,各剂量组与阴性对照组比较,GJIC水平再次下降,差异均具有统计学意义(P<0.05),并具有一定剂量-反应关系。结论煤焦油能够抑制GJIC功能。Objective To investigate the effects of coal tar on gap junction intercellular communication (GJIC)in NIH/3T3 cells of mice. Methods Taking NIH/3T3 cells of mice as the target cells, the effects of coal tar on GJIC in NIH/3T3 cells were examined by the scrape-loading and dye transfer(SLDT)technique. Results The NIH/3T3 cells were exposed to coal tar of 0.1, 1.5 and 22.5 μg/ml for 6 hours respectively. Compared with the control group, the level of GJIC in the highest dose group lowered when exposed for 6 hours, the difference was significant (P〈0. 05). As the exposure prolonged to 12 hours, the levels of GJIC in NIH/3T3 cells in all dosage groups further lowered, all the differences were significant (P〈0. 05) showing dose-response relationship. Conclusions The coal tar might inhibit the GJIC function of NIH/3T3 cells.
