摘  要:本文研究的主要目的是建立大肠杆菌可溶性蛋白质组的双向电泳技术,以等点聚焦为第一向,SDS-PAGE为第二向进行双向电泳,并对样品处理方式、蛋白上样量和染色方法等关键因素和环节进行了比较研究,通过实验条件的筛选和优化获得了较满意的双向电泳图谱。采取的方法具有较高的分辨率和重复性,为进一步研究其蛋白质组学奠定了基础,成功地用于了本科实验教学中。The main purpose of the study is to establish groups of soluble protein in Escherichia coli by two- dimensional electrophoresis technology, the first phase is isoelectrofocusing, and the second electrophoresis is SDS-PAGE. The effect of sample handling, protein staining methods on the sample volume and other key factors and segments were discussed and studied comparatively. The more satisfactory two-dimensional electrophoresis maps could be gained through screening and optimization of experimental conditions. This Methods in our experiment has a high resolution and repeatability, which has laid a foundation for study of proteomics and been used successfully in undergraduate teaching experiment.
